目的:观察脾气虚证大鼠肠神经Cajal间质细胞(ICC)间信号转导通路的表达改变和香砂六君子汤的治疗作用。方法:将100只Wistar大鼠随机分为对照组、模型组、自然恢复组和香砂六君子汤治疗组,采用免疫荧光双标记染色、激光扫描共聚焦显微镜观察模型组大鼠小肠肠神经-ICC信号转导通路损伤和香砂六君子汤的修复作用。结果:脾气虚证大鼠小肠ICC计数和荧光强度(IOD)值明显降低,网络样结构遭到破坏,PKA/PKC蛋白荧光强度IOD值明显降低。结论:香砂六君子汤能够修复脾气虚证大鼠小肠Cajal间质细胞和信号转导通路的损伤,改善胃肠运动障碍,治疗脾气虚证。
目的:觀察脾氣虛證大鼠腸神經Cajal間質細胞(ICC)間信號轉導通路的錶達改變和香砂六君子湯的治療作用。方法:將100隻Wistar大鼠隨機分為對照組、模型組、自然恢複組和香砂六君子湯治療組,採用免疫熒光雙標記染色、激光掃描共聚焦顯微鏡觀察模型組大鼠小腸腸神經-ICC信號轉導通路損傷和香砂六君子湯的脩複作用。結果:脾氣虛證大鼠小腸ICC計數和熒光彊度(IOD)值明顯降低,網絡樣結構遭到破壞,PKA/PKC蛋白熒光彊度IOD值明顯降低。結論:香砂六君子湯能夠脩複脾氣虛證大鼠小腸Cajal間質細胞和信號轉導通路的損傷,改善胃腸運動障礙,治療脾氣虛證。
목적:관찰비기허증대서장신경Cajal간질세포(ICC)간신호전도통로적표체개변화향사륙군자탕적치료작용。방법:장100지Wistar대서수궤분위대조조、모형조、자연회복조화향사륙군자탕치료조,채용면역형광쌍표기염색、격광소묘공취초현미경관찰모형조대서소장장신경-ICC신호전도통로손상화향사륙군자탕적수복작용。결과:비기허증대서소장ICC계수화형광강도(IOD)치명현강저,망락양결구조도파배,PKA/PKC단백형광강도IOD치명현강저。결론:향사륙군자탕능구수복비기허증대서소장Cajal간질세포화신호전도통로적손상,개선위장운동장애,치료비기허증。
Objective To observe the expression changes of signal transduction pathway of enteric nerne system-interstitial cell (ENS-ICC) and the therapeutic effects of Xiang Sha Liu Jun Zi decoction(XSLJZD) in rats with deficiency of Spleen-Qi syndreme (DSQS). Methods One hundred Wistar rats were randomly divid-ed into control group, DSQS group, recovery group and XSLJZD group. The signal transduction pathway of ENS-ICC of the gastrointestinal tract was observed by using immunohistochemical double-staining with whole-amount preparation technique and confocal laser scanning microcopy. Results The IOD of ICC of intes-tine in DSQS group was significantly lower than that in control group(P<0.01), the ICC network was significant-ly disrupted. The IOD of ICC of intestine in the recovery group was slightly better than that in DSQS group(P<0.01), the ICC network was slightly recovered. The IOD of ICC of intestine in XSLJZD treatment group was sig-nificantly increased(P<0.01). After treatment with XSLJZD, the ICC network was significantly recovered. The IOD of protein kinase A(PKA)/protein kinase C(PKC) in DSQS group was significantly decreased(P<0.01). The signal transduction pathway was destroyed. The IOD of protein kinase A(PKA)/protein kinase C(PKC) in the re-covery group was slightly recovered(P<0.01). While the IOD of protein kinase A(PKA)/protein kinase C(PKC) in XSLJZD treatment group was significantly increased. The signal transduction pathway was significantly recov-ered. Conclusion XSLJZD can improve the gastrointestinal motility in DSQS and the mechanism may be re-lated to the repairing effect on the distribution and the connection of Signal Transduction Pathway of ENS-ICC in rat with DSQS.
