以改良Trizol试剂法从富含多糖的甜玉米幼穗中提取高质量、完整的总RNA,利用实时荧光定量PCR对高温胁迫下粤甜13雌穗发育的8个差异表达基因进行分析。结果表明,8个基因分为上调表达和下调表达,实时荧光定量PCR和测序法基因表达谱检测的基因上调或下调表达的趋势一致,上调表达基因ZM2G058057和下调表达基因ZM2G059964、ZM2G044670相对表达量较高,可能在高温胁迫影响甜玉米幼雌穗发育过程中起更重要的作用。
以改良Trizol試劑法從富含多糖的甜玉米幼穗中提取高質量、完整的總RNA,利用實時熒光定量PCR對高溫脅迫下粵甜13雌穗髮育的8箇差異錶達基因進行分析。結果錶明,8箇基因分為上調錶達和下調錶達,實時熒光定量PCR和測序法基因錶達譜檢測的基因上調或下調錶達的趨勢一緻,上調錶達基因ZM2G058057和下調錶達基因ZM2G059964、ZM2G044670相對錶達量較高,可能在高溫脅迫影響甜玉米幼雌穗髮育過程中起更重要的作用。
이개량Trizol시제법종부함다당적첨옥미유수중제취고질량、완정적총RNA,이용실시형광정량PCR대고온협박하월첨13자수발육적8개차이표체기인진행분석。결과표명,8개기인분위상조표체화하조표체,실시형광정량PCR화측서법기인표체보검측적기인상조혹하조표체적추세일치,상조표체기인ZM2G058057화하조표체기인ZM2G059964、ZM2G044670상대표체량교고,가능재고온협박영향첨옥미유자수발육과정중기경중요적작용。
An improved Trizol method was used for extracting the total RNA from polysaccharide-rich young ears of sweet corn. The extract total RNA was high quality and integrity. Real-time quantitative PCR was used to investi-gate 8 genes expressions in ear development of cultivar Yuetian13 under heat stress. The results indicated that 8 genes were divided into up- or down-regulated types, and their expression patterns by Real-time quantitative PCR were basically consistent with the digital gene expression data. Three of 8 genes, up-regulated expression gene ZM2G058057, down-regulated expression genes ZM2G059964 and ZM2G044670 had higher relative expression lev-els in ears, which may play very important roles in ears development under heat stress.
