为建立一套最适合叶用莴苣(Lactuca sativa L.)基因表达量研究的荧光定量PCR技术体系,以叶用莴苣S24为试材,采用SYBR GreenⅠ法,利用叶用莴苣内参基因的2对引物组合18SR1和18SR2,分别在10μL和20μL反应体系条件下进行扩增反应。结果表明,分别在10μL和20μL反应体系条件下内参基因引物组合18SR1的扩增曲线、标准曲线以及溶解曲线等指标都优于内参基因引物组合18SR2;2对内参基因引物组合18SR1和18SR2在10μL反应体系中的扩增曲线、标准曲线以及溶解曲线等指标也均优于20μL反应体系。因此叶用莴苣荧光定量PCR内参基因利用18SR1引物组合,反应体系采用10μL比较合理。
為建立一套最適閤葉用萵苣(Lactuca sativa L.)基因錶達量研究的熒光定量PCR技術體繫,以葉用萵苣S24為試材,採用SYBR GreenⅠ法,利用葉用萵苣內參基因的2對引物組閤18SR1和18SR2,分彆在10μL和20μL反應體繫條件下進行擴增反應。結果錶明,分彆在10μL和20μL反應體繫條件下內參基因引物組閤18SR1的擴增麯線、標準麯線以及溶解麯線等指標都優于內參基因引物組閤18SR2;2對內參基因引物組閤18SR1和18SR2在10μL反應體繫中的擴增麯線、標準麯線以及溶解麯線等指標也均優于20μL反應體繫。因此葉用萵苣熒光定量PCR內參基因利用18SR1引物組閤,反應體繫採用10μL比較閤理。
위건립일투최괄합협용와거(Lactuca sativa L.)기인표체량연구적형광정량PCR기술체계,이협용와거S24위시재,채용SYBR GreenⅠ법,이용협용와거내삼기인적2대인물조합18SR1화18SR2,분별재10μL화20μL반응체계조건하진행확증반응。결과표명,분별재10μL화20μL반응체계조건하내삼기인인물조합18SR1적확증곡선、표준곡선이급용해곡선등지표도우우내삼기인인물조합18SR2;2대내삼기인인물조합18SR1화18SR2재10μL반응체계중적확증곡선、표준곡선이급용해곡선등지표야균우우20μL반응체계。인차협용와거형광정량PCR내삼기인이용18SR1인물조합,반응체계채용10μL비교합리。
In order to establish a method real-time PCR for studying gene expression quantity of Lactuca sative L., with leaves of Lactuca sativa L. (S24) as material, SYBR GreenⅠmethod had been used, and two pairs of Lactuca sativa L. actin gene primers 18SR1 and 18SR2 were used in 10 μL and 20 μL reaction system conditions amplification reaction. The results showed that primers 18SR1 amplification curve, standard curve and dissolution curve related index were better than primers 18SR2 in 10 μL and 20 μL reaction system conditions. 18SR1 and 18SR2 primers combinations in 10 μL reaction system in amplification curve, standard curve and dissolution curve related index were better than 20 μL reaction system. So Lactuca sativa L. 18SR1 primers combination and 10 μL reaction system using was more reasonable.
