目的:探讨检测原发性胆汁性肝硬化(PBC)患者外周血 CD4+ CD25+ Foxp3+调节性 T 淋巴细胞的水平与意义。方法采用流式细胞术检测32例 PBC 患者和23例健康体检者外周血 CD4+ CD25+ Foxp3+调节性 T 淋巴细胞亚群比例;采用实时荧光定量聚酶链反应(PCR)技术检测人叉头型基因 p3(Foxp3)mRNA 表达的水平。结果PBC 患者外周血 CD4+ CD25+ Foxp3+调节性 T 淋巴细胞明显低于对照组(P <0.05);Foxp3的 mRNA 水平差异无统计学意义(P >0.05)。结论PBC 患者外周血 CD4+ CD25+ Foxp3+调节性 T 淋巴细胞减少,免疫抑制功能减弱,提示 CD4+ CD25+ Foxp3+调节性 T 淋巴细胞在 PBC 的免疫发病机制中可能起重要作用,对评估 PBC 患者细胞免疫状态和预后有重要价值。

목적:탐토검측원발성담즙성간경화(PBC)환자외주혈 CD4+ CD25+ Foxp3+조절성 T 림파세포적수평여의의。방법채용류식세포술검측32례 PBC 환자화23례건강체검자외주혈 CD4+ CD25+ Foxp3+조절성 T 림파세포아군비례;채용실시형광정량취매련반응(PCR)기술검측인차두형기인 p3(Foxp3)mRNA 표체적수평。결과PBC 환자외주혈 CD4+ CD25+ Foxp3+조절성 T 림파세포명현저우대조조(P <0.05);Foxp3적 mRNA 수평차이무통계학의의(P >0.05)。결론PBC 환자외주혈 CD4+ CD25+ Foxp3+조절성 T 림파세포감소,면역억제공능감약,제시 CD4+ CD25+ Foxp3+조절성 T 림파세포재 PBC 적면역발병궤제중가능기중요작용,대평고 PBC 환자세포면역상태화예후유중요개치。
Objective To investigate the roles of CD4+ CD25 + Foxp3 + regulatory T cells (Treg)in the primary bili-ary cirrhosis patients.Methods The flow cytometry was used to evaluated the population of CD4+ CD25 + T cells in the peripheral blood of the PBS patients.Forkhead/winged helix transcription factor(Foxp3)mRNA expression levels of each group were detected by real-time-fluorescence quantitative retroverse trascriptase polymerase chain reaction(RT-PCR).Results The primary biliary cirrhosis group showed significantly higher levels of CD4+ CD25 + Foxp3 + regula-tory T cells in peripheral blood than those of control group(P <0.05),and there were no difference for the mRNA level of Foxp3+ in Treg.Conclusion The expression of CD4+ CD25 + Foxp3 + T cells was decreased in patients with primary biliary cirrhosis(PBC)and weakened immune suppression function,suggesting that they may play an important role in immunopathogenesis of PBC,the level of blood Treg can refelect the cell immune state of PBC and of clinical value to assess the prognosis.
