生物法是处理聚乙烯醇(PVA)废水的最主要的方法之一,关键是筛选获得高效PVA降解菌。采集的土壤样品经85℃水浴及富集培养,并采用I2-KI及硼酸染色法进行初筛,初筛菌株通过测定PVA降解酶酶活的方法进行复筛。初筛得到PVA降解菌9株,复筛得到4株PVA降解酶酶活力较高的菌株,其中菌株1-21的酶活最高为1.136 U/mL。通过对该菌株形态学观察、生理生化试验及16S rDNA 序列分析,初步鉴定该菌株属于为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),菌株1-21与相应标准菌株BCRC 11601的16s rDNA序列相似度达99.77%。通过对PVA高效降解菌株的筛选可以为用生物方法高效处理PVA废水提供优势菌株并降低处理废水的成本。

Biological method was one of the main methods to deal with polyvinyl alcohol (PVA) wastewater, and the key was to screen for efficient PVA degradation bacteria. The collected soil samples was handled with 85℃ water bath and enrichment culture, the I2-KI and boric acid staining method was used for initial screening, the screening strain was through the method of testing PVA-degrading enzymes for multiple screening. 9 PVA degradation strains were obtained in the initial screening, 4 PVA degradation strains were obtained in the multiple screening. PVA-degrading enzymes was high, the highest PVA-degrading enzymes of the strain 1-21 was up to 1.136 U/mL. Through the strain morphological observation, physiological and biochemical experiments and 16 s rDNA sequence analysis, strain was identified as a Bacillus amyloliquefaciens, the similarity of the 16S rDNA sequences between strain 1-21 and the type strain BCRC 11601 of Bacillus amyloliquefaciens was close to 99.77%. Through the PVA of high efficiency degradation bacterium screening could provide advantage strain which could be used as a biological method efficient processing PVA wastewater and reduce the cost of wastewater treatment.
