Epilepsy%GnT - Vb%Dystroglycans
目的:探讨难治性癫痫细胞模型( Sombati 癫痫细胞模型)中 N -乙酰氨基葡萄糖转移酶 Vb (GnT - Vb)和肌营养不良蛋白聚糖(dystroglycan)基因的表达变化及意义。方法取新生24 h 内 SD 乳鼠,分离其双侧海马进行原代海马神经元培养。将培养至第14天的海马神经元分为对照组和模型组,模型组利用低镁细胞外液培养3 h 制备 Sombati 癫痫细胞模型。通过实时定量聚合酶链式反应(real - time qPCR)方法测定对照组和模型组造模后第1天和第4天 GnT - Vb 和 dystroglycan 的基因表达水平。结果造模后第1天,模型组 GnT - Vb 基因表达水平低于对照组(P <0.05);而两组 dystroglycan 基因表达水平间无差异(P >0.05)。造模后第4天,模型组 GnT - Vb、dystrogly-can 基因表达水平均高于对照组(P <0.05)。结论 GnT - Vb 和 dystroglycan 基因在 Sombati 癫痫细胞模型中表达呈增高趋势,并呈现一定时间的动态变化;GnT - Vb 和 dystroglycan 基因可能同时参与了难治性癫痫的形成过程。

목적:탐토난치성전간세포모형( Sombati 전간세포모형)중 N -을선안기포도당전이매 Vb (GnT - Vb)화기영양불량단백취당(dystroglycan)기인적표체변화급의의。방법취신생24 h 내 SD 유서,분리기쌍측해마진행원대해마신경원배양。장배양지제14천적해마신경원분위대조조화모형조,모형조이용저미세포외액배양3 h 제비 Sombati 전간세포모형。통과실시정량취합매련식반응(real - time qPCR)방법측정대조조화모형조조모후제1천화제4천 GnT - Vb 화 dystroglycan 적기인표체수평。결과조모후제1천,모형조 GnT - Vb 기인표체수평저우대조조(P <0.05);이량조 dystroglycan 기인표체수평간무차이(P >0.05)。조모후제4천,모형조 GnT - Vb、dystrogly-can 기인표체수평균고우대조조(P <0.05)。결론 GnT - Vb 화 dystroglycan 기인재 Sombati 전간세포모형중표체정증고추세,병정현일정시간적동태변화;GnT - Vb 화 dystroglycan 기인가능동시삼여료난치성전간적형성과정。
Objective To investigate the gene expression changes of GnT - Vb and dystroglycan in the Sombati cell culture model of refractory epilepsy. Methods The hippocampal neurons which were isolated from bilateral hippocamp of new-born SD rats within 24 hours of birth,were cultured in vitro. On the 14th day,the neurons were divided into control group and experimental group. The experimental group was cultured with low Mg2 + extracellular fluid for 3 hours to establish Sombati cell culture model. The real - time qPCR was used to detect the mRNA expression levels of GnT - Vb and dystroglycan in control group and experimental group on the first day and the fourth day. Results On the first day after model establishment,the mRNA expression level of GnT - Vb in experimental group was significantly lower than that in control group(P < 0. 05). There was no significant difference in mRNA expression level of dystroglycan between two groups(P > 0. 05). On the fourth day after model establishment,mRNA expression levels of GnT - Vb and dystroglycan in experimental group were significantly higher than those in control group(P < 0. 05). Conclusion The increasing trend of mRNA expression levels of GnT - Vb and dystroglycan can be observed in the Sombati cell culture model,and the expression levels change in a specific time period. The mRNA of GnT - Vb and dystrogly may participate in the formation process of refractory epilepsy.
