为了筛选适用于玉米耐酸性土壤鉴定的分子标记,根据已报道与玉米耐铝性有关的2个主要QTL(ZmM1和 ZmM2)的 Indel 标记连锁图与 IBM2 Neighbors Map 的相关区段连锁图,将 ZmM1和ZmM2基因附近的分子标记进行整合,选择距离 ZmM1和 ZmM2基因较近的4个 SSR 标记,共计6个标记用于4个自交系(其中1个为耐酸性土壤自交系)的耐铝性分子标记多态性检测。结果表明:在 ZmM1位点,QCL8007-5和 QCL8002-11不具备 CML530的耐酸性等位,而 QCL8003-2具备 CML530的耐酸性土壤等位。在 ZmM2位点,QCL8003-2和 QCL8002-11没有 CML530的耐酸性,而 QCL8007-5具有 CML530的耐酸性等位。在 umc2309和 umc1468两位点,QCL8007-5、QCL8003-2和 QCL8002-11不具有 CML530的耐酸性土壤等位。在 umc2068和 umc2578两位点,QCL8007-5、QCL8003-2和 QCL8002-11具有 CML530的耐酸性土壤等位。综合考虑标记的多态性和遗传连锁关系,在对 ZmM1和 ZmM2位点进行分子标记辅助选择时,宜选择标记 ZmMATE1、ZmMATE2和 umc1468进行检测。
為瞭篩選適用于玉米耐痠性土壤鑒定的分子標記,根據已報道與玉米耐鋁性有關的2箇主要QTL(ZmM1和 ZmM2)的 Indel 標記連鎖圖與 IBM2 Neighbors Map 的相關區段連鎖圖,將 ZmM1和ZmM2基因附近的分子標記進行整閤,選擇距離 ZmM1和 ZmM2基因較近的4箇 SSR 標記,共計6箇標記用于4箇自交繫(其中1箇為耐痠性土壤自交繫)的耐鋁性分子標記多態性檢測。結果錶明:在 ZmM1位點,QCL8007-5和 QCL8002-11不具備 CML530的耐痠性等位,而 QCL8003-2具備 CML530的耐痠性土壤等位。在 ZmM2位點,QCL8003-2和 QCL8002-11沒有 CML530的耐痠性,而 QCL8007-5具有 CML530的耐痠性等位。在 umc2309和 umc1468兩位點,QCL8007-5、QCL8003-2和 QCL8002-11不具有 CML530的耐痠性土壤等位。在 umc2068和 umc2578兩位點,QCL8007-5、QCL8003-2和 QCL8002-11具有 CML530的耐痠性土壤等位。綜閤攷慮標記的多態性和遺傳連鎖關繫,在對 ZmM1和 ZmM2位點進行分子標記輔助選擇時,宜選擇標記 ZmMATE1、ZmMATE2和 umc1468進行檢測。

The purpose of the study was to screen the molecular markers for acid soil tolerance in maize.According to the reported Indel marker linkage maps of two major Al tolerant QTL (ZmM1 and ZmM2)and IBM2 Neighbors Maps of their bins,two integrated bin linkage maps were constructed through realignment of markers flanking them.Then,4 SSR markers flanking ZmM1 and ZmM2 genes and two Indel markers of ZmMATE1 and ZmMATE2 were used to detect polymorphisms in 4 inbred lines including an acid soil tolerant line of CML530.The results showed that QCL8007-5 and QCL8002-11 didn't have any acid soil tolerant allele of CML530 at ZmM1 locus,while QCL8003-2 had;QCL8003-2 and QCL8002-11 didn't possess any acid soil tolerant allele of CML530 at ZmM2 locus,while QCL8007-5 possessed.At the two loci of umc2309 and umc1468,all three lines of QCL8007-5、QCL8003-2 and QCL8002-11 didn't bear any acid soil tolerant allele of CML530;However,at the two loci of umc2068 and umc2578,those three lines carried. Therefore, considering the polymorphism and genetic linkage relationship of the 6 markers,the three markers of ZmMATE1,ZmMATE2 and umc1468 should be suitable for molecular marker-assisted selection at the ZmM1 and ZmM2 loci.
