以千层金(Melaleuca bracteata F. Muell.)的幼嫩叶片为外植体,探讨不同灭菌方法对外植体的表面灭菌效果及不同植物生长调节剂组合与浓度配比对千层金叶片愈伤组织诱导及分化的影响.结果表明:75%酒精浸泡30s+1%次氯酸钠溶液灭菌15min 的组合为千层金叶片外植体适宜的灭菌方法;千层金叶片在附加2,4-D、NAA 和6-BA 的 MS 培养基上均能诱导出愈伤组织,其中诱导愈伤组织的最佳培养基为MS+NAA2mg/L+6-BA0.1mg/L,诱导率可达100%,诱导不定芽分化的最佳培养基为 MS+6-BA1mg/L+NAA0.1mg/L,分化率为19.8%.

Using the tender leaves of Melaleuca bracteata F. Muell. as explants, the effect of different disinfection methods on explants and the influence of plant growth regulators on callus induction and differentiation were studied. The results showed that the disinfection method of 75% alcohol to deal with for 30 s and 1% NaClO to deal with for 15 min was suitable for tender leaves. Callus could be induced on MS medium supplemented with different combinations of 2,4-D,NAA and 6-BA. The optimum medium for callus induction was MS+ NAA2mg/L +6-BA 0.1mg/L with 100% of callus induction rate. The optimum medium for bud differentiation of callus was MS+6-BA 1mg/L +NAA0.1mg/L with 19.8% of differentiation rate.
