目的观察甲基化转移酶抑制剂地西他滨对2型糖尿病KKAy小鼠血糖和体重的影响,探讨其可能机制。方法将20只2型糖尿病KKAy小鼠按随机数字表法分为两组,实验组10只使用地西他滨治疗,剂量为15 mg? m-2? d-1,对照组10只使用生理盐水治疗,记录两组治疗前后的血糖、体重。结果治疗结束后,对照组小鼠的血糖和体重均增加(P<0.05),实验组体重增加(P<0.05),而血糖治疗前后差异无统计学意义(P>0.05)。治疗后实验组血糖、体重及增加幅度均低于对照组(P<0.05)。结论甲基化转移酶抑制剂能缓解2型糖尿病小鼠血糖和肥胖的恶化,其机制可能与能量代谢通路和胰岛素信号的重要基因发生了去甲基化有关。

목적관찰갑기화전이매억제제지서타빈대2형당뇨병KKAy소서혈당화체중적영향,탐토기가능궤제。방법장20지2형당뇨병KKAy소서안수궤수자표법분위량조,실험조10지사용지서타빈치료,제량위15 mg? m-2? d-1,대조조10지사용생리염수치료,기록량조치료전후적혈당、체중。결과치료결속후,대조조소서적혈당화체중균증가(P<0.05),실험조체중증가(P<0.05),이혈당치료전후차이무통계학의의(P>0.05)。치료후실험조혈당、체중급증가폭도균저우대조조(P<0.05)。결론갑기화전이매억제제능완해2형당뇨병소서혈당화비반적악화,기궤제가능여능량대사통로화이도소신호적중요기인발생료거갑기화유관。
Objective To observe the effect of methyltransferases (DNMTs) inhibitor on blood glucose and body weight of KKAy mice with type 2 diabetes mellitus(T2DM),and explore the potential mechanism .Methods Twenty KKAy mice with T2DM were randomly divided into 2 groups.The mice in the treatment group(n=10) were treated with decitabine at a dose of 15 mg? m-2? d-1 while the mice in the control group (n=10) were treated with normal saline.Blood glucose level and body weight of the mice in both groups were recorded before and after the treatment . Results Blood glucose level and body weight of the mice in the control group increased after treatment (P<0.05),while in the treatment group,body weight increased after treatment (P<0.05) but blood glucose level showed no significant difference before and after treatment(P>0.05).After treatment,blood glucose level,body weight and their increase in the treatment group were significantly lower than those in the control group (P<0.05).Conclusion DNMTs inhibitor may delay the deterioration of blood glucose and body weight in T 2DM diabetes mice.The demethylation in some important genes which relates with energy metabolism and insulin signals may be the potential mechanism .
