目的:观察塞来昔布体外实验效应及相关影响因素,探索其体外药物作用的相关机制。方法利用MTT 比色法测定塞来昔布对多种细胞系、药物作用不同时间以及与细胞短时间接触后产生的抑制率;通过测定血红蛋白的吸光度来反应不同条件下药物引起的溶血作用;观察不同培养基血清浓度、药物配置后放置不同时间、超声及滤膜过滤等物理处理方式对药物体外作用的影响。结果体外培养基条件下,塞来昔布可同时抑制肿瘤细胞系及正常细胞系的生长,药物的这种作用在与药物短暂接触后即会产生,且药物抑制作用会随作用时间的延长而逐渐增强;较高浓度药物可在短时间内引起红细胞胞膜破裂,这种作用可在4℃的环境下中止,恢复温度后红细胞再次破裂;培养基中血清浓度、药物配置后放置时间以及超声和滤膜过滤等因素均可明显影响药物的体外抑制作用。结论培养基中未溶解的游离形态药物是塞来昔布体外效应的作用主体,其通过快速黏附于细胞膜系统而产生一系列特殊的实验现象,故可影响药物游离形式的方式均可明显影响药物的体外效应。

목적:관찰새래석포체외실험효응급상관영향인소,탐색기체외약물작용적상관궤제。방법이용MTT 비색법측정새래석포대다충세포계、약물작용불동시간이급여세포단시간접촉후산생적억제솔;통과측정혈홍단백적흡광도래반응불동조건하약물인기적용혈작용;관찰불동배양기혈청농도、약물배치후방치불동시간、초성급려막과려등물리처리방식대약물체외작용적영향。결과체외배양기조건하,새래석포가동시억제종류세포계급정상세포계적생장,약물적저충작용재여약물단잠접촉후즉회산생,차약물억제작용회수작용시간적연장이축점증강;교고농도약물가재단시간내인기홍세포포막파렬,저충작용가재4℃적배경하중지,회복온도후홍세포재차파렬;배양기중혈청농도、약물배치후방치시간이급초성화려막과려등인소균가명현영향약물적체외억제작용。결론배양기중미용해적유리형태약물시새래석포체외효응적작용주체,기통과쾌속점부우세포막계통이산생일계렬특수적실험현상,고가영향약물유리형식적방식균가명현영향약물적체외효응。
Objective To explore the mechanism of celecoxib in vitro action by analyzing celecoxib′s various special experiments in vitro and related influential factors. Methods MTT assay was used to detect the effect of celecoxib on a varie-ty of cell lines and it was also be used to detect the inhibition rates at different time and the rates after contacting with the drug for a short time;Hemolysis caused by the drugs under different conditions is measured by the absorbance of hemoglobin;Some physical methods have been used to evaluate the related influence factors in vitro,such as the different serum concentration, placed several hours after drug configuration,ultrasound processing and filter with the 0. 22μm filtration. Results Celecoxib can inhibit the growth of tumor cell lines and normal cell lines,this effect can arise after a short time contacting and can en-hance by prolonging the action time. High concentration of drug can cause erythrocyte hemolysis immediately,this effect can be suspended in 4℃,and it will burst again when recovery to 37℃ . Different serum concentration,placed several hours after drug configuration as well as the factors such as ultrasonic processing and filter with 0. 22μm filtration can significantly affect the in-hibition rates of the drug. Conclusion The free-undissolved form of celecoxib in vitro culture medium play the major role in vitro action,and it produce a series of special experimental phenomena through the rapid adhesion on the cell membrane sys-tem,those which can influence the free form of the drug can significantly change the drug action in vitro.
