Radix astragali%Hippocampal DG%Taurine%Learning and memory%rats
目的:观察黄芪水提液对大鼠海马齿状回(DG)区氨基酸物质的含量以及学习记忆能力的影响。方法Wistar 系雄性大鼠24只,随机分为对照组、黄芪小剂量组及黄芪大剂量组。利用脑部微量透析法和高效液相色谱法,观察大鼠海马 DG 区细胞外液中的六种氨基酸的含量,包括天冬氨酸、谷氨酸、谷氨酰胺、甘氨酸、牛磺酸和丙氨酸,其后,用相应溶液灌胃3周,再次观察海马 DG 区六种氨基酸的含量,并利用穿梭箱观察大鼠的学习记忆能力。结果对照组海马 DG 区的六种氨基酸在灌胃前后无明显变化,而黄芪大、小剂量组海马 DG 区的牛磺酸含量灌胃后出现明显的增加(均为 P <0.05);在5天的穿梭箱训练过程中,黄芪大、小剂量组的每天的训练所需时间逐渐缩短,与对照组相比有显著性差异(均为 P <0.05)。结论黄芪增强大鼠的学习记忆能力,其作用机制可能与增加海马 DG 区牛磺酸含量有关。

목적:관찰황기수제액대대서해마치상회(DG)구안기산물질적함량이급학습기억능력적영향。방법Wistar 계웅성대서24지,수궤분위대조조、황기소제량조급황기대제량조。이용뇌부미량투석법화고효액상색보법,관찰대서해마 DG 구세포외액중적륙충안기산적함량,포괄천동안산、곡안산、곡안선알、감안산、우광산화병안산,기후,용상응용액관위3주,재차관찰해마 DG 구륙충안기산적함량,병이용천사상관찰대서적학습기억능력。결과대조조해마 DG 구적륙충안기산재관위전후무명현변화,이황기대、소제량조해마 DG 구적우광산함량관위후출현명현적증가(균위 P <0.05);재5천적천사상훈련과정중,황기대、소제량조적매천적훈련소수시간축점축단,여대조조상비유현저성차이(균위 P <0.05)。결론황기증강대서적학습기억능력,기작용궤제가능여증가해마 DG 구우광산함량유관。
Objective To investigate the effects of Radix astragali on extracelluar levels of amino acids in the hipp-ocampal dentate gyrus (DG)region in rats.Methods Totally 24 Wistar rats were randomly divided into three groups, control group,Radix astragali low dose group and Radix astragali large dose group.The rats were intragastric adminis-trated with Radix astragali or saline 3w,and then learing and memory were detected by shuttle box.The extracellular concentrations of aspirate (Asp),glutamate (Glu),glutamine (Gln),glycine (Gly),taurine (Tau)and alanine (Ala)in the hippocampal DG were measured by microdialysis and high performance liquid chromatography (HPLC)techniques before and after of intragastric administration.Results Compared with control group,the extracellular concentrations of Tau in the hippocampal DG was significantly increased in the Radix astragali groups (P <0.05).Moreover,the Radix astragali groups showed significantly shorter training time compared with control group (P <0.05).Conclusion Radix astragali enhance the ability of learning and memory in rats,which maybe related to increase in Tau concentrations in the hippocampal DG region.
