目的:观察茵栀黄注射液对急性肝衰竭大鼠细胞凋亡的影响。方法:30只SD大鼠随机分为正常组、模型组和茵栀黄组。茵栀黄组大鼠予0.2g/(kg·d)茵栀黄注射液腹腔注射,连续给药7d,末次给药后1h,除正常组外其余两组给予 D-氨基半乳糖(D-Ga1N)和内毒素脂多糖(LPS)腹腔注射制备大鼠急性肝衰竭模型。造模后6h,取大鼠肝组织,采用原位末端标记(TUNEL)法检测肝细胞凋亡情况,并以免疫组化法(SP法)检测各组大鼠肝组织Bcl-2和Bax蛋白表达情况。结果:与正常组比较,模型组肝组织中Bcl-2蛋白表达明显下降,肝细胞凋亡指数和Bax蛋白表达均明显升高(P均<0.01);与模型组比较,茵栀黄组肝细胞凋亡指数和Bax蛋白表达均明显下调,而Bcl-2蛋白表达明显上调(P均<0.05)。结论:茵栀黄注射液对急性肝衰竭大鼠肝细胞凋亡有保护作用,其作用机制可能与上调Bcl-2蛋白表达和下调Bax蛋白表达有关。

목적:관찰인치황주사액대급성간쇠갈대서세포조망적영향。방법:30지SD대서수궤분위정상조、모형조화인치황조。인치황조대서여0.2g/(kg·d)인치황주사액복강주사,련속급약7d,말차급약후1h,제정상조외기여량조급여 D-안기반유당(D-Ga1N)화내독소지다당(LPS)복강주사제비대서급성간쇠갈모형。조모후6h,취대서간조직,채용원위말단표기(TUNEL)법검측간세포조망정황,병이면역조화법(SP법)검측각조대서간조직Bcl-2화Bax단백표체정황。결과:여정상조비교,모형조간조직중Bcl-2단백표체명현하강,간세포조망지수화Bax단백표체균명현승고(P균<0.01);여모형조비교,인치황조간세포조망지수화Bax단백표체균명현하조,이Bcl-2단백표체명현상조(P균<0.05)。결론:인치황주사액대급성간쇠갈대서간세포조망유보호작용,기작용궤제가능여상조Bcl-2단백표체화하조Bax단백표체유관。
Objective:To investigate effects on the apoptosis of rats with acute liver failure by the Yinzhihuang inject. Methods:30 SD rats were divided into normal group, model group and Yinzhihuang group, the Yinzhihuang group was given the Yinzhihuang injection 0.2g/(kg·d), and 7 days consecutively, 1 hour after the last time, both group were given D-Ga1N and LPS except the normal group, in order to make the model, 6h later, take the liver tissue of rats In situ end labeling (TUNEL) assay hepatocyte apoptosis, and Bcl-2 and Bax protein expression and detection of liver tissue by immunohistochemical method (SP). Results:Compared with the normal group, Bcl-2 protein expression was significantly decreased in the liver tissue of the model group, the the liver apoptotic index and Bax protein expression were significantly increased (P<0.01). Compared with the model group, the group of Yinzhihuang hepatocyte apoptosis index and Bax protein expression were significantly down-regulated Bcl-2 protein expression was significantly up-regulated (P<0.05). Conclusion:The Yinzhihuang injection have protective effect on the apoptosis of rats with acute liver failure, its mechanism of action may be related to the increase in Bcl-2 and Bax down.
