在已构建好的蜡梅花 cDNA文库及 EST分析的基础上,随机克隆测序得到一个蜡梅 FKBP 基因,将其命名为CpFKBP,Genebank登录号:JQ337962.该基因cDNA长为482 bp,具有一个339 bp的ORF,编码一个113个氨基酸的多肽,预测等电点为8.48,理论分子量为12.08 KD.在大肠杆菌中获得并纯化了重组的 CpFKBP 蛋白质,并进一步对重组蛋白进行PPIase活性分析,结果显示CpFKBP重组蛋白有较高的PPIase酶活性.
在已構建好的蠟梅花 cDNA文庫及 EST分析的基礎上,隨機剋隆測序得到一箇蠟梅 FKBP 基因,將其命名為CpFKBP,Genebank登錄號:JQ337962.該基因cDNA長為482 bp,具有一箇339 bp的ORF,編碼一箇113箇氨基痠的多肽,預測等電點為8.48,理論分子量為12.08 KD.在大腸桿菌中穫得併純化瞭重組的 CpFKBP 蛋白質,併進一步對重組蛋白進行PPIase活性分析,結果顯示CpFKBP重組蛋白有較高的PPIase酶活性.

A FKBP gene has been obtained by randomly sequencing the cDNA library of Chimonanthus praecox flowers,which is named as CpFKBP (GenBank accession No:JQ337962).The 482 bp cDNA of CpFKBP contains a 339 bp open reading frame (ORF)encoding a 113-amino-acid polypeptide with a cal-culated molecular mass of 12.08 KD and the theoretical pI of 8.48.The recombinant CpFKBP with the fused TRX and His Tags at it N-terminus has been purified by the His-binding purification Kit.The analy-sis of PPIase activity shows that the fusion protein of CpFKBP has a high PPIase activity.This study has facilitated the research of the FKBP genes in different species,the application of immuno-suppressants and understanding the whole immunophilin family.
