目的：探讨替罗非班对氧化型低密度脂蛋白（ox-LDL）刺激下 THP-1源性巨噬细胞 CD40和CD40配体（CD40L）表达的影响，探讨该类药物的抗炎及稳定斑块的作用机制。方法向体外培养的 THP-1细胞中加入 ox-LDL（80 mg／L）共同孵育24 h，然后与替罗非班共同孵育不同时间（分别为12、24、48 h），应用流式细胞技术检测 CD40和 CD40L 在细胞上的表达。采用反转录-聚合酶链反应（RT-PCR）检测 CD40和 CD40L mRNA 表达。结果替罗非班既可下调细胞表面 CD40和 CD40L 的表达，又能使 CD40和 CD40L mRNA 表达下降，其阻断作用呈时间依赖性（P ＜0.05）。结论替罗非班可以拮抗巨噬细胞 CD40和 CD40L 表达，并呈时间依赖性，这种作用可能是其减轻动脉粥样斑块炎症及稳定斑块的机制之一。
목적：탐토체라비반대양화형저밀도지단백（ox-LDL）자격하 THP-1원성거서세포 CD40화CD40배체（CD40L）표체적영향，탐토해류약물적항염급은정반괴적작용궤제。방법향체외배양적 THP-1세포중가입 ox-LDL（80 mg／L）공동부육24 h，연후여체라비반공동부육불동시간（분별위12、24、48 h），응용류식세포기술검측 CD40화 CD40L 재세포상적표체。채용반전록-취합매련반응（RT-PCR）검측 CD40화 CD40L mRNA 표체。결과체라비반기가하조세포표면 CD40화 CD40L 적표체，우능사 CD40화 CD40L mRNA 표체하강，기조단작용정시간의뢰성（P ＜0.05）。결론체라비반가이길항거서세포 CD40화 CD40L 표체，병정시간의뢰성，저충작용가능시기감경동맥죽양반괴염증급은정반괴적궤제지일。
Objective To investigate the effects of tirofiban on expression of CD40,CD40L in THP-1 cells, which is stimulated by ox-LDL,and to investigate the drug resistance to anti-inflammatory and plaque stability mechanism.Methods THP-1 cells were induced by fixed concentration of ox-LDL (80 mg/L)for 24 hours.Then with tirofiban incubated together at different times (respectively,12,24,48 h),the CD40 and CD40L expression on the macrophage were detected by flow cytometric analysis and CD40 and CD40L mRNA were detected by RT-PCR.Results Tirofiban can down-regulate not only the expression of CD40 and CD40L,but also the mRNA lev-els of CD40 and CD40L in cultured macrophage.In addition,the tirofiban inhibited them by time dependence(P <0.05).Conclusion Tirofiban inhibited the expression of CD40,CD40L concentration dependently in THP-1 cells,which is stimulated by ox-LDL.The antagonism is related to its anti-inflammatory and plaque stability.
