Artesunate%Fibroblast%Apoptosis%Scar
目的:探讨青蒿琥酯(Art)抑制人成纤维细胞(Fb)增殖的作用及机制。方法取人瘢痕组织,经体外培养及鉴定Fb后,将不同浓度的青蒿琥酯溶液作用于体外培养的成纤维细胞。于倒置显微镜及电镜观察其形态学变化,采用四氮唑盐比色分析法(MTT法)及流式细胞仪观察细胞凋亡,计算增殖抑制率和凋亡指数。结果青蒿琥酯对人瘢痕成纤维有明显抑制作用,且呈浓度依赖性。浓度为240、120 mg/L的青蒿琥酯的凋亡率和坏死率较对照组高(P<0.01)。组织学观察试验组见成纤维细胞呈椭圆性,胞质颗粒增多,甚至可见细胞核浓缩、核碎裂等现象。结论青蒿琥酯可抑制人成纤维细胞的增殖,具体机制需进一步探讨。

목적:탐토청호호지(Art)억제인성섬유세포(Fb)증식적작용급궤제。방법취인반흔조직,경체외배양급감정Fb후,장불동농도적청호호지용액작용우체외배양적성섬유세포。우도치현미경급전경관찰기형태학변화,채용사담서염비색분석법(MTT법)급류식세포의관찰세포조망,계산증식억제솔화조망지수。결과청호호지대인반흔성섬유유명현억제작용,차정농도의뢰성。농도위240、120 mg/L적청호호지적조망솔화배사솔교대조조고(P<0.01)。조직학관찰시험조견성섬유세포정타원성,포질과립증다,심지가견세포핵농축、핵쇄렬등현상。결론청호호지가억제인성섬유세포적증식,구체궤제수진일보탐토。
Objective Investigate the effects of artesunate(Art) inhibit fibroblast(Fb) proliferation effect and mechanism. Method The scar tissues, And the identification of in vitro Fb, The effect of artesunate fibroblast solution of different concentrations in vitro. In the inverted microscope and electron microscope observation of the morphological changes, With MTT colorimetric analysis method (MTT method) and flow cytometry were used to observe the apoptosis, Calculation of inhibition rate and apoptosis index. Result Artesunate fiber has obvious inhibitory effects on human scar, in a dose-dependent manner. Concentration of 240, 120 mg/L of artesunate apoptosis rate and necrosis rate than the control group (P<0.01). Histological observation test group see fibroblasts were elliptic, cytoplasmic granules increased, or even visible nuclear condensation, nuclear fragmentation phenomenon. Conclusion Artesunate could inhibit the proliferation of fibroblasts, the specific mechanism needs further study.
