目的 探究KB细胞被单纯疱疹病毒Ⅰ型(herpes simplex virus,-1,HSV-1)体外感染的途径及方式.方法 取患者的疱疹液、唾液、咽喉漱洗液等标本,采用乳鼠脑内接种的方法培养分离HSV,从而获取研究所需的HSV-1,将获取的该病毒进行KB细胞感染,采用倒置显微镜鉴定病毒感染的细胞形态学改变,同时采用聚合酶链式反应技术(PCR)检测感染细胞中的HSV-1病毒的核酸,并将感染后的上清液收集起来进行乳鼠脑细胞接种,采用倒置显微镜鉴定病毒感染的细胞形态学改变.结果 在倒置显微镜下进行观察,并未发现典型细胞病变,而上清液感染后的乳鼠脑细胞则发现了典型的细胞病变,采用PCR法检测HSV-1感染KB细胞提取的DNA,可见HSV-1病毒核酸.结论 单纯疱疹病毒Ⅰ型能对KB细胞进行直接感染.

목적 탐구KB세포피단순포진병독Ⅰ형(herpes simplex virus,-1,HSV-1)체외감염적도경급방식.방법 취환자적포진액、타액、인후수세액등표본,채용유서뇌내접충적방법배양분리HSV,종이획취연구소수적HSV-1,장획취적해병독진행KB세포감염,채용도치현미경감정병독감염적세포형태학개변,동시채용취합매련식반응기술(PCR)검측감염세포중적HSV-1병독적핵산,병장감염후적상청액수집기래진행유서뇌세포접충,채용도치현미경감정병독감염적세포형태학개변.결과 재도치현미경하진행관찰,병미발현전형세포병변,이상청액감염후적유서뇌세포칙발현료전형적세포병변,채용PCR법검측HSV-1감염KB세포제취적DNA,가견HSV-1병독핵산.결론 단순포진병독Ⅰ형능대KB세포진행직접감염.
Objective To explore the approach and mode of the KB cells infected by the herpes simplex virus type Ⅰ (HSV-1) in vitro.Methods Collecting the herpes fluid,saliva and throat rinsing fluid from the patients.In order to gain the HSV-1 required for the research,HSV was isolated by intracerebral injection to sucking mice.KB cells infected with the virus were identified by the inverted microscope to observe the morphological changes of the cell ; the viral DNA was also detected using polymerase chain reaction (PCR) in the infected cells.And the supernatant was collected after infection for the intracerebral injection to sucking mice,and identified the morphological changes of the infected cells by the inverted microscope.Results The typical cytopathy was not been observed under the inverted microscope in infected KB cells,but the typical cytopathy was found in the brain cells of suckling mice intracerebral injected with the supernatant,the viral DNA was detected in the KB cells infected with HSV-1 by PCR.Conclusions The herpes simplex virus type Ⅰ can directly infect the cells,and we must pay more attention to it in order to reduce the damage of human oral epithelial cell caused by HSV-1.
