利用常压室温等离子体射流诱变和紫外照射对夫西地酸生产菌株进行复合诱变,得到3株产量明显提高的突变菌株,3株菌的平均发酵效价较出发菌株提高14%;然后,采用均匀设计实验对其中发酵效价提高最多的 AU-37菌株的发酵培养基碳、氮源进行了优化,得到适合该菌株的发酵培养基优化配方为:糊精1.86%、花生饼粉2.5%、蛋白胨0.3%、氯化铵0.5%、七水合硫酸镁0.3%、硫酸钾0.6%、碳酸钙1%、pH 值7.2,在优化的发酵培养基中,AU-37菌株摇瓶发酵效价较出发菌株提高33.8%,为工业化生产奠定了基础。

Fusidic acid-producing strain FA-30 was mutated by ARTP-UV composite mutagenesis,three high-efficient producing strains were obtained by screening,which was increased in fermentation potency by 14% over the original strain.The carbon source and nitrogen source of the fermentation medium for strain AU-37 with the highest fermentation potency were optimized through the uniform design experiment.The optimum fermentation medium was as follows:dextrin 1.86%,peanut cake powder 2.5%,peptone 0.3%,NH 4 Cl 0.5%, MgSO4 ·7H2 O 0.3%,K2 SO4 0.6%,CaCO3 1%,pH value 7.2,and the fermentation potency of strain AU-37 cultured in the above medium reached 33.8% over that of the original strain.
