高效液相色谱法测定非发酵豆制品中人工合成色素的含量。样品用石油醚去脂,用硫酸和钨酸钠溶液除去蛋白质,聚酰胺粉吸附色素,洗涤后解吸,纯化后测定。采用 Thermo ODS-2 HYPERSIL(150 mm ×4.6 mm ×5μm)色谱柱,流动相为0.02 mol/ L 乙酸铵-甲醇,梯度洗脱,流速为1.0 mL/ min,柱温30℃,检测波长427,485 nm,进样量20μL。结果表明,柠檬黄进样量在4~160 ng(Y =3079.0X -1980.4,r =0.9998)、日落黄进样量在7~280 ng(Y =2537.1X -1507.1,r =1.0000)范围内呈现良好的线性,柠檬黄、日落黄平均加样回收率为100.8%,100.4%。该方法能快速检测、操作简便且灵敏度高。

To develop a quantitative method for artificial synthetic pigment in non-fermented bean products by HPLC. Used petroleum ether degreasing method,removal of the protein in solution of sulfuric acid and sodium tungstate,polyamide adsorption pigments,determine after washing with purified analysis. The col-umn for Thermo ODS-2 HYPERSIL(150 mm × 4. 6 mm × 5 μm),the mobile phase was 0. 02 mol/ L am-monium acetate methanol,gradient elution;the flow rate was 1. 0 mL/ min;column temperature was 30 ℃;the detection wavelength:427,485 nm;sample 20 μL. Results showed that the lemon yellow in the 4 ~160 ng(Y =3 079. 0X -1 980. 4,r =0. 999 8),sunset yellow in the 7 ~ 280 ng(Y = 2 537. 1X - 1 507. 1, r = 1. 000 0)range showed a good linear,lemon yellow,sunset yellow average recovery rate of 100. 8% , 100. 4% . The rapid detection method has the advantages of simple operation,high sensitivity.
