目的:观察异种(猪)脱细胞真皮基质(Xeno-ADM)对大鼠深Ⅱ度烧伤创面中肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、白细胞介素-1(IL-1)表达的影响。方法取70只6~8周健康雄性Wister大鼠于背部建立深Ⅱ度烧伤模型,并随机分为治疗组和对照组,各组35只。治疗组大鼠烧伤创面采用Xeno-ADM覆盖包扎,对照组大鼠烧伤创面采用传统外用聚维酮碘换药包扎。按分组情况分别于伤后1、3、5、7、10、14、21 d每组各取5个大鼠,沿创缘取创面组织标本,采用免疫组织化学染色法和免疫印迹等技术检测创面愈合过程中炎症介质 TNF-a、IL-8、IL-1的表达水平,采用SSPS 11.0统计软件对统计学数据进行处理,组间比较采用配对t检验。结果免疫组化提示:两组IL-8,TNF-a 和IL-1均在第1天开始表达,第5天达高峰,第7~21天表达水平逐渐下降。其中,伤后治疗组的TNF-a、IL-8表达均低于对照组,且上述差异均有统计学意义(P<0.05)。而IL-1的表达在第5天到第7天实验组明显高于对照组,且差异均有统计学意义(P<0.05)。免疫印记显示,上述检测因子在两组中的表达规律及组间差异均与免疫组化结果相一致。结论 Xeno-ADM可有效抑制创面IL-8、TNF-a的表达和促进IL-1的表达,从而减轻炎症反应,促进创面愈合。

목적:관찰이충(저)탈세포진피기질(Xeno-ADM)대대서심Ⅱ도소상창면중종류배사인자-α(TNF-α)、백세포개소-8(IL-8)、백세포개소-1(IL-1)표체적영향。방법취70지6~8주건강웅성Wister대서우배부건립심Ⅱ도소상모형,병수궤분위치료조화대조조,각조35지。치료조대서소상창면채용Xeno-ADM복개포찰,대조조대서소상창면채용전통외용취유동전환약포찰。안분조정황분별우상후1、3、5、7、10、14、21 d매조각취5개대서,연창연취창면조직표본,채용면역조직화학염색법화면역인적등기술검측창면유합과정중염증개질 TNF-a、IL-8、IL-1적표체수평,채용SSPS 11.0통계연건대통계학수거진행처리,조간비교채용배대t검험。결과면역조화제시:량조IL-8,TNF-a 화IL-1균재제1천개시표체,제5천체고봉,제7~21천표체수평축점하강。기중,상후치료조적TNF-a、IL-8표체균저우대조조,차상술차이균유통계학의의(P<0.05)。이IL-1적표체재제5천도제7천실험조명현고우대조조,차차이균유통계학의의(P<0.05)。면역인기현시,상술검측인자재량조중적표체규률급조간차이균여면역조화결과상일치。결론 Xeno-ADM가유효억제창면IL-8、TNF-a적표체화촉진IL-1적표체,종이감경염증반응,촉진창면유합。
Objective To discuss the influence of xenogenic acellular dermal matrix on theexpression of Tumor Necrosis Factor-alpha, Interleukin-8, Interleukin-1 in wound healing of deep back of 70 male Wister rats.Rats were randomly divided into experimental group and control group,eachgroup only 35.The rats in experimental group were treated with Xeno-ADM,while the rats in control groupwere treated by changing the dressing in a traditional way.Samples were collected at 1 d,3 d,5 d,7 d,10 d,14 d and 21 d post-scalding.Immunohistochemistry and Western Blotting were respectively conductedto examine the expression of Tumor Necrosis Factor-alpha, Interleukin-8 and Interleukin-1 in bothexperimental and control groups.SPSS11.0 statistical software was used to process the data.Comparisionbetween groups was made using the paired t test.Results Trough Western Blotting,Interleukin-8,TumorNecrosis Factor-alpha and Interleukin-1 were observed on the 1st day.Their expression reached a peak onthe fifth day and gradually declined from the 7th to 21th days.The expression of Interleukin-8 and TumorNecrosis Factor-alpha in experimental group was lower significantly than that in control group during21 days.The expression level of Interleukin-1 in experimental group was higher significantly than that incontrol group from the 5th to 7th days.The above differences of the expression between experimental groupand control group had statistical significance(P <0.05).Immunohistochemistry revealed the expression anddifferences of IL-1,IL-8 and TNF-a in the both experimental and control groups were consistent with theresults of Western Blotting.Conclusions Xeno-ADM can indicate active cells proliferation through down-regulating the expression of Interleukin-8 and Tumor Necrosis Factor-alpha and up-regulating the expressionof Interleukin-1,accelerating burned wound healing,shorten the treatment courses.
