目的:观察高血糖对肝纤维化大鼠血清中转化生长因子-β1(TGF-β1)、结缔组织生长因子(CTGF)含量的影响,探讨高血糖与肝纤维化之间的相关性。方法将40只SD大鼠按照体重随机分为4组,每组10只,分别为对照组(C组)、高血糖组(H组)、肝纤维化组(M组)、高血糖+肝纤维化组(H+M组),其中用四氯化碳造肝纤维化模型,用链脲佐菌素诱导出现高血糖,8周后将动物处死,取血,采用酶联免疫分析法( ELISA法)检测4组动物血清中TGF-β1、CTGF的含量,采用实时定量RT-PCR检测4组动物血清中TGF-β1、CTGFmRNA的表达。结果 TGF-β1、CTGF含量及mR-NA表达在H组与C组相比无显著差异( P >0.05),M组及H+M组血清中上述指标明显高于C组( P <0.05),而H+M组血清中上述指标升高更明显( P <0.05);且TGF-β1与CTGF之间具有明显的正相关( r =0.812,P <0.05)。结论高血糖能够增加肝纤维化大鼠血清中TGF-β1及CTGF的表达及含量,且TGF-β1与CTGF含量与肝纤维化程度呈正相关,可以作为评价肝纤维化的重要指标。

목적:관찰고혈당대간섬유화대서혈청중전화생장인자-β1(TGF-β1)、결체조직생장인자(CTGF)함량적영향,탐토고혈당여간섬유화지간적상관성。방법장40지SD대서안조체중수궤분위4조,매조10지,분별위대조조(C조)、고혈당조(H조)、간섬유화조(M조)、고혈당+간섬유화조(H+M조),기중용사록화탄조간섬유화모형,용련뇨좌균소유도출현고혈당,8주후장동물처사,취혈,채용매련면역분석법( ELISA법)검측4조동물혈청중TGF-β1、CTGF적함량,채용실시정량RT-PCR검측4조동물혈청중TGF-β1、CTGFmRNA적표체。결과 TGF-β1、CTGF함량급mR-NA표체재H조여C조상비무현저차이( P >0.05),M조급H+M조혈청중상술지표명현고우C조( P <0.05),이H+M조혈청중상술지표승고경명현( P <0.05);차TGF-β1여CTGF지간구유명현적정상관( r =0.812,P <0.05)。결론고혈당능구증가간섬유화대서혈청중TGF-β1급CTGF적표체급함량,차TGF-β1여CTGF함량여간섬유화정도정정상관,가이작위평개간섬유화적중요지표。
Objective ToobservetheeffectsofhyperglycemiaontheexpressionofTGF-β1,CTGF,andtoexplorehyperglycemiaand correlationbetweenhepaticfibrosis.Methods 40SDratswererandomlydividedintofourgroupsaccordingtobodyweight,n=10,thecontrol group( C group),hyperglycemia group( H group),liver fibrosis group( M),hyperglycemia + liver fibrosis group( H+M group). The liver fi-brosis was made with carbon tetrachloride liver fibrosis model,with the emergence of streptozotocin-induced hyperglycemia. All the animals were killed after eight weeks. The TGF-β1 and CTGF levels were detected by ELISA. The TGF-β1 and CTGF mRNA expression were detected by real-timequantitativeRT-PCR.Results TGF-β1,CTGFlevelsandmRNAexpressioningroupCandHgroupdidnotchangesignificantly ( P >0. 05). Serum levels of these parameters of M and H + M group were significantly higher than that in group C( P <0. 05). While the ser-um these indicators of H + M group increased more significantly( P <0. 05). TGF-β1 and CTGF has a significant positive correlation( r =0.812,P<0.05).Conclusion HighbloodsugarcanincreaseserumliverfibrosisTGF-β1andCTGFexpressionandcontent.TheTGF-β1 and CTGF content and degree of liver fibrosis has a positive correlation between the evaluation of liver fibrosis. These indexes can be used as an im-portant indicator.
