目的:观察柿叶黄酮对同型半胱氨酸诱导的小胶质细胞表达白介素-1茁(IL-1茁)、白介素-6(IL-6)和肿瘤坏死因子-琢(TNF-琢)的影响,并与银杏叶提取物EGb761进行比较。方法将体外培养的小鼠小胶质细胞(BV-2细胞)分成空白对照组、同型半胱氨酸组(Hcy)、Hcy+EGb761组(100mg/L)和Hcy+低、中、高(12.5、25、50滋g/mL)剂量柿叶黄酮组,培养72h。应用RT-qPCR方法评价各组IL-1茁、IL-6和TNF-琢的mRNA表达,酶联免疫吸附法(ELISA)测定培养上清液中上述细胞因子的蛋白浓度。结果与空白对照组比较,同型半胱氨酸组细胞内IL-1茁、IL-6和TNF-琢的mRNA表达和细胞培养上清液中蛋白浓度明显增加(P<0.05);与同型半胱氨酸组比较,Hcy+EGb761组和Hcy+低、中、高剂量柿叶黄酮组各细胞因子mRNA表达及培养上清液中蛋白浓度均降低(P<0.05),尤其是高剂量柿叶黄酮组结果与Hcy+EGb761组作用相似。结论柿叶黄酮能抑制同型半胱氨酸诱导小胶质细胞表达IL-1茁、IL-6和TNF-琢,在一定浓度条件下其作用不亚于银杏叶提取物。

목적:관찰시협황동대동형반광안산유도적소효질세포표체백개소-1촬(IL-1촬)、백개소-6(IL-6)화종류배사인자-탁(TNF-탁)적영향,병여은행협제취물EGb761진행비교。방법장체외배양적소서소효질세포(BV-2세포)분성공백대조조、동형반광안산조(Hcy)、Hcy+EGb761조(100mg/L)화Hcy+저、중、고(12.5、25、50자g/mL)제량시협황동조,배양72h。응용RT-qPCR방법평개각조IL-1촬、IL-6화TNF-탁적mRNA표체,매련면역흡부법(ELISA)측정배양상청액중상술세포인자적단백농도。결과여공백대조조비교,동형반광안산조세포내IL-1촬、IL-6화TNF-탁적mRNA표체화세포배양상청액중단백농도명현증가(P<0.05);여동형반광안산조비교,Hcy+EGb761조화Hcy+저、중、고제량시협황동조각세포인자mRNA표체급배양상청액중단백농도균강저(P<0.05),우기시고제량시협황동조결과여Hcy+EGb761조작용상사。결론시협황동능억제동형반광안산유도소효질세포표체IL-1촬、IL-6화TNF-탁,재일정농도조건하기작용불아우은행협제취물。
Objective To evaluate the effect of flavone from diospyros kaki leaves on the expression of inter-leukin-1β(IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in microgia(BV-2 cells) induced by homocysteine,and compared its effect with that of EGb761. Methods The BV-2 cells incubated in vitro were di vided into six groups:blank control group,homocysteine(Hcy) group,EGb761 group(100mg/L) and flavone from diospyros kaki groups of different concentration(12.5,25,50μg/mL). After 72h incubation,the mRNA expression of IL-1β,IL-6 and TNF-α was assessed by RT-qPCR and the protein of cytokines in supernatant was detected by ELISA. Results The mRNA expression and supernatant protein of IL-1β,IL-6,TNF-α in Hcy group were signifi-cantly higher than those of control group(P<0.05),but were significantly decreased in EGb761 group and flavone from diospyros kaki groups compared with those of Hcy group(P<0.05). Especially,both the mRNA expression and supernatant protein in higher concentration flavone from diospyros kaki leaf group were comparable to those in EGb761 group. Conclusion Flavone from diospyros kaki leaves can inhibit the expression of IL-1β,IL-6 and TNF-α in cultured BV-2 cells induced by Hcy. In a certain concentration,it was no less effective than EGb761.
