目的:探讨生长激素( GH)对心肌成纤维细胞( CFs)增殖的影响及其作用机制。方法以不同浓度的重组人生长激素(rhGH)(100、200、500、1000 ng/ml)和磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)抑制剂LY294002(10μmol/L)单独或同时作用CFs 48 h后,采用MTT法检测细胞增殖,Wertern Blot检测PI3K/Akt信号途径中Akt和磷酸化Akt ( p-Akt )的蛋白水平的变化。结果 rhGH呈浓度依赖性的促进CFs的增殖;rhGH增加p-Akt的蛋白水平;LY294002可阻断rhGH诱导的CFs的增殖。结论生长激素可通过激活PI3K/Akt信号通路促进心肌成纤维细胞的增殖。

목적:탐토생장격소( GH)대심기성섬유세포( CFs)증식적영향급기작용궤제。방법이불동농도적중조인생장격소(rhGH)(100、200、500、1000 ng/ml)화린지선기순3-격매/단백격매B(PI3K/Akt)억제제LY294002(10μmol/L)단독혹동시작용CFs 48 h후,채용MTT법검측세포증식,Wertern Blot검측PI3K/Akt신호도경중Akt화린산화Akt ( p-Akt )적단백수평적변화。결과 rhGH정농도의뢰성적촉진CFs적증식;rhGH증가p-Akt적단백수평;LY294002가조단rhGH유도적CFs적증식。결론생장격소가통과격활PI3K/Akt신호통로촉진심기성섬유세포적증식。
Objective To investigate the effect of growth hormone ( GH) on the proliferation of cardiac fibroblasts (CFs) and its mechanism.Methods CFs were treated by different concentrations of recombinant human growth hormone (rhGH) (100,200,500,1000 ng/ml) with or without phosphatidylinositol-3-kinase/protein kinase B ( PI3K/Akt) inhibitor LY294002 (10μmol/L) for 48 hours.MTT assay was used to determine the proliferation of CFs .Western Blot was used to detect the protein levels of Akt and phosphorylated Akt ( p-Akt) in PI3K/Akt signaling pathway .Results rhGH promoted the proliferation of CFs in a concentration-dependent manner .rhGH increased the protein level of p-Akt.LY294002 could depress the rhGH-induced proliferation of CFs .Conclusion GH promotes the proliferation of CFs by activation of PI3K/Akt signaling pathway .
