目的：对大容量样本中丙型肝炎病毒（HCV）进行浓缩提取，采用自行设计引物进行实时 HCV 荧光定量检测，验证病毒浓缩的效率以及病毒浓缩对检测的影响。方法采用 PEG 法高速离心进行病毒浓缩，一步法逆转录荧光定量 PCR 验证病毒浓缩效果。结果该方案用于 HCV RNA 检测，能得到良好的扩增曲线，不同病毒载量样本病毒浓缩的效率均在70%以上。结论大容量样本 HCV 病毒浓缩可提高病毒荧光 PCR 检测的灵敏度。
목적：대대용량양본중병형간염병독（HCV）진행농축제취，채용자행설계인물진행실시 HCV 형광정량검측，험증병독농축적효솔이급병독농축대검측적영향。방법채용 PEG 법고속리심진행병독농축，일보법역전록형광정량 PCR 험증병독농축효과。결과해방안용우 HCV RNA 검측，능득도량호적확증곡선，불동병독재량양본병독농축적효솔균재70%이상。결론대용량양본 HCV 병독농축가제고병독형광 PCR 검측적령민도。
Objective The objectives of the present study are to first concentrate and extract Hepatitis C Virus(HCV)from large sam-ples,and then to identify the efficiency of the virus concentration and its effects on detection by reverse transcription qRT - PCR with custom de-signed primer. Methods HCV virus concentrated by high speed centrifugation with PEG,and quantitatively determined by one - step RT - PCR. Results This method can be used for HCV RNA test. The results indicated more than 70% concentrating efficiency in samples with different viral load. Conclusion The concentration of HCV virus in large samples can improve the sensitivity of the virus detection by RT - PCR.
