目的:检测白细胞介素( IL)-18在非小细胞肺癌高、低转移株表达的差异。方法采用半定量 RT-PCR及 Western 印迹技术,检测IL-18在非小细胞肺癌高、低转移株的差异表达;构建IL-18 cDNA的重组质粒,同时把IL-18基因导入非小细胞肺癌高、低转移株,观察 IL-18对肿瘤细胞体外转移能力的影响。结果与非小细胞肺癌低转移株相比, IL-18 mRNA 和蛋白均只能在非小细胞肺癌高转移株检测到,表达量分别为(5.41±1.20)、(3.98±2.01)。将IL-18基因导入非小细胞肺癌低转移株后,转染细胞的体外迁移能力显著上调,为空白对照组的3倍左右。结论非小细胞肺癌高转移株可组成性表达IL-18,且IL-18表达促进非小细胞肺癌的转移。

목적:검측백세포개소( IL)-18재비소세포폐암고、저전이주표체적차이。방법채용반정량 RT-PCR급 Western 인적기술,검측IL-18재비소세포폐암고、저전이주적차이표체;구건IL-18 cDNA적중조질립,동시파IL-18기인도입비소세포폐암고、저전이주,관찰 IL-18대종류세포체외전이능력적영향。결과여비소세포폐암저전이주상비, IL-18 mRNA 화단백균지능재비소세포폐암고전이주검측도,표체량분별위(5.41±1.20)、(3.98±2.01)。장IL-18기인도입비소세포폐암저전이주후,전염세포적체외천이능력현저상조,위공백대조조적3배좌우。결론비소세포폐암고전이주가조성성표체IL-18,차IL-18표체촉진비소세포폐암적전이。
Objective To compare interleukin ( IL )-18 expression between high and low metastatic plant in non-small cell lung cancer(NSCLC).Methods Semi-quantitative RT-PCR and Western blot analysis were used to compare IL-18 expressions between high and low metastatic plant in NSCLC;IL-18 cDNA recombinant plasmid was published , at the same time the IL-18 genes were leaded in NSCLC with high and low strain .IL-18 effects on tumor cells in vitro metastasis ability were observed .Results Compared with NSCLC with low transfer plant, IL-18 mRNA and protein were only detected in high metastatic strains of NSCLC , expression quantity were (5.41±1.20) (3.98±2.01) respectively, after introducing IL-18 genes in NSCLC into low metastatic plant , metastasis cells in vitro migration ability to rise significantly , about 3 times as the blank control group .Conclusions With IL-18 expression in high metastatic plants of NSCLC , IL-18 expression promotes the metastasis of NSCLC cells .
