目的:通过对髓源性神经干细胞与运动神经元的 miRNA 表达谱的测定,确定运动神经元中高表达的miRNA 和低表达 miRNA 的个数,研究这种方法的可行性。方法采用 Applied Biosystems 公司的 TagMan 低密度阵列(TLDA)技术实现对二者的 miRNA 表达谱的技术检测,并通过 PCR 生产 U6 snRNA 的扩增曲线,对二者的差异性进行比较。结果运动神经元中高表达的 miRNA 共有60个,低表达的 miRNA 共有35个,髓源性神经干细胞与运动神经元两种细胞的扩增曲线呈现出明显的“S”形曲线。对于4种具有代表性的 TLDA 筛选出来的4种具有差异性的 miRNA 进行表达检测,并对检测结果进行定量结果分析,并进行相关线性比较,经过模拟曲线得出线性相关 r =0.978,P =0.017,显示相关性好。结论髓源性神经干细胞与运动神经元两种细胞的扩增曲线呈现出明显的“ S”形曲线,可见二者的表达性差异。TLDA 技术在髓源性神经干细胞与运动神经元的 miRNA 表达谱检测中发挥重要的作用,值得推广和应用。

Objective To determine the marrow - derived neural stem cells and motor neurons miRNA expression profiling,to determine the number of miRNA and miRNA expression lower motor neurons in the high expression to study the feasibility of this approach. Methods This method of this experimental study is produced by the company Applied Biosystems TLDA technology,which was used to detect miRNA expression profiles. By PCR amplification curves,U6 snRNA of production was used to compare the differences between the two group. Results For the marrow - derived neural stem cells and motor neurons miRNA expression profiling studies,we identified miRNA expression in motor neurons of a total of 60,miRNA total of 35 low expression. A representative for the four kinds TLDA screening out of four kinds of miRNA expression were de-tected. The test results for quantitative analysis of results was associated with linear comparison. The curve showed simulation of linear correlation r = 0. 978. Conclusion Marrow - derived neural stem cells and amplification curves of two motor neuron cells showing a clear "S" shaped curve, showing expression differences between them. TLDA technology miRNA marrow - derived neural stem cells and expression profiling of motor neu-rons play an important role in the detection. This method should be worthy of promotion and application.
