目的:探讨长脉冲 Nd:YAG 1064 nm 激光对临床分离不同表型红色毛癣菌形态学的影响。方法长脉冲Nd:YAG 1064 nm 激光分别以200 J/ cm2、400 J/ cm2、600 J/ cm2能量体外照射含有等菌量的红色毛癣菌 Y 菌落(绒毛型)和红色毛癣菌 N 菌落(沟纹型),将激光照射前后的菌落制备成标本,分别在扫描电镜(SEM)及透射电镜(TEM)观察其形态结构变化。结果对绒毛型 T. rubrum Y 菌落,随着激光照射能量的增加,与对照组相比,绒毛型 T. rubrum Y 菌丝的形态结构破坏明显。表现为 SEM 下,菌丝结构变得粗糙、表面有凹陷、局部形成膨大,皱瘪非常明显;TEM 下,菌体双层细胞壁及细胞器结构模糊或破坏,胞浆内可见脂滴和空泡变性,髓样小体及蛋白凝固体(为凋亡的表现),甚至菌体崩解、坏死。对沟纹型 T. rubrum N 菌落,随着激光照射能量的增加,与对照组相比,沟纹型 T. rubrum N 菌丝的形态结构破坏不显著。表现为 SEM 下菌丝结构变得弯曲、粗糙,表面有少量凹陷,菌丝局部膨大,无明显皱瘪;TEM 下,菌体双层细胞壁及细胞器结构模糊或不连续,胞浆内可见脂滴和空泡变性,无凋亡及坏死表现。结论绒毛型红色毛癣菌 Y 菌落在激光照射后,其菌丝形态发生明显变化;而沟纹型红色毛癣菌 N 菌落在激光照射后,其菌丝形态变化相对较小。

목적:탐토장맥충 Nd:YAG 1064 nm 격광대림상분리불동표형홍색모선균형태학적영향。방법장맥충Nd:YAG 1064 nm 격광분별이200 J/ cm2、400 J/ cm2、600 J/ cm2능량체외조사함유등균량적홍색모선균 Y 균락(융모형)화홍색모선균 N 균락(구문형),장격광조사전후적균락제비성표본,분별재소묘전경(SEM)급투사전경(TEM)관찰기형태결구변화。결과대융모형 T. rubrum Y 균락,수착격광조사능량적증가,여대조조상비,융모형 T. rubrum Y 균사적형태결구파배명현。표현위 SEM 하,균사결구변득조조、표면유요함、국부형성팽대,추별비상명현;TEM 하,균체쌍층세포벽급세포기결구모호혹파배,포장내가견지적화공포변성,수양소체급단백응고체(위조망적표현),심지균체붕해、배사。대구문형 T. rubrum N 균락,수착격광조사능량적증가,여대조조상비,구문형 T. rubrum N 균사적형태결구파배불현저。표현위 SEM 하균사결구변득만곡、조조,표면유소량요함,균사국부팽대,무명현추별;TEM 하,균체쌍층세포벽급세포기결구모호혹불련속,포장내가견지적화공포변성,무조망급배사표현。결론융모형홍색모선균 Y 균락재격광조사후,기균사형태발생명현변화;이구문형홍색모선균 N 균락재격광조사후,기균사형태변화상대교소。
Objective To explore the effect of long pulse Nd:YAG 1064 nm laser irradiation on morphological changes of Trichophyton rubrum Y(T. rubrum Y)and Trichophyton rubrum N(T. rubrum N). Methods Equal number of colonies of T. rubrum Y(phenotype vil-lous)was irradiated in vitro by long pulse Nd:YAG 1 064 nm laser with different energy(0 J/ cm2 ,200 J/ cm2 ,400 J/ cm2 ,600 J/ cm2 )and 0 J/ cm2 for control group. The same procedure had been done for colonies of T. rubrum N(phenotype furrowed)again. The above fungal samples were prepared before and after laser treatment in order to observe the morphological changes of T. rubrum Y and T. rubrum N by scanning electron microscopy(SEM)and transmission electron microscopy(TEM). Results For villous colonies of T. rubrum Y,followed by upgrading laser en-ergy treatment,the morphological feature of hyphae of T. rubrum Y had been severely destroyed. Under SEM,the structure of hyphae of villous T. rubrum Y became more rough,clearly bending,obviously shrunken and deflated. Furthermore many craters on the surface had been observed un-der SEM compared to control group. Under TEM,the structure of two layers of cell wall and cell organelle of T. rubrum Y became indistinct or dis-continuous,and lipid droplet,vacuolar degeneration,myeloid body and albuminous degeneration(these latter two performance indicate apoptosis) could be observed in Cysoplasm. Finally,irreversible necrosis happened. For furrowed colonies of T. rubrum N,followed by increasing laser irra-diation,there were similar changes including rough surface with lightly bending,short swollen of hyphae. However,no obvious shrunken and de-flated hyphae had been seen under SEM. TEM screening showed fuzzy or deformed two layers of cell wall,obscure or dissolved cell organelle, some lipid droplet and vacuolar degeneration were observed in T. rubrum N. No apoptosis or necrosis had been observed. Conclusion The obvi-ously morphological changes were observed in villous colonies of T. rubrum Y after laser treatment. In contrast with these changes,the slightly morphological changes were observed in furrowed colonies of T. rubrum N after laser treatment.
