目的:探讨 cAMP反应原件结合蛋白(cAMP response element binding protein,CREB)和CXC趋化因子受体2(CXC chemokine receptor 2,CXCR2)在肺鳞癌和腺癌组织中的表达及与肺癌组织分化程度的关系。方法随机抽取肺癌手术切除组织标本49例,其中鳞癌22例,腺癌27例,高-中分化肺癌组织19例,低分化肺癌组织30例。采用免疫组化Elivision法分别检测CREB和CXCR2在分化程度不同的肺癌组织中的表达情况,根据肺癌组织细胞的染色率和染色强度进行判断,分析各组之间的表达及相关性。以20例肺良性病变组织标本作为阴性对照,所有患者均有完善的术前检查和术后病理诊断,其中支气管扩张7例,炎性假瘤9例,支气管囊肿4例。结果 CREB阳性信号主要位于胞核和胞浆,CXCR2阳性信号主要位于胞浆和胞膜;肺癌组织中CREB和CXCR2的表达水平明显高于肺良性病变组织(P<0.01);低分化肺癌组织中CREB和CXCR2的表达明显高于高-中分化(P<0.05);CREB和 CXCR2的表达存在相关性。结论 CXCR2和 CREB在肺癌组织中的表达水平明显增高;肺癌的分化程度越低,肺癌的危险性越高,CXCR2和CREB的表达水平越强。

목적:탐토 cAMP반응원건결합단백(cAMP response element binding protein,CREB)화CXC추화인자수체2(CXC chemokine receptor 2,CXCR2)재폐린암화선암조직중적표체급여폐암조직분화정도적관계。방법수궤추취폐암수술절제조직표본49례,기중린암22례,선암27례,고-중분화폐암조직19례,저분화폐암조직30례。채용면역조화Elivision법분별검측CREB화CXCR2재분화정도불동적폐암조직중적표체정황,근거폐암조직세포적염색솔화염색강도진행판단,분석각조지간적표체급상관성。이20례폐량성병변조직표본작위음성대조,소유환자균유완선적술전검사화술후병리진단,기중지기관확장7례,염성가류9례,지기관낭종4례。결과 CREB양성신호주요위우포핵화포장,CXCR2양성신호주요위우포장화포막;폐암조직중CREB화CXCR2적표체수평명현고우폐량성병변조직(P<0.01);저분화폐암조직중CREB화CXCR2적표체명현고우고-중분화(P<0.05);CREB화 CXCR2적표체존재상관성。결론 CXCR2화 CREB재폐암조직중적표체수평명현증고;폐암적분화정도월저,폐암적위험성월고,CXCR2화CREB적표체수평월강。
Obj ective To investigate the expression of CREB and CXCR2 in lung squamous cell carci-noma and adenocarcinoma,and to evaluate their relationship with the histological differentiation grades. Methods Surgical samples of 49 cases of lung cancer were randomly selected,of which 22 were squamous cell carcinoma and 27 adenocarcinoma,19 were well-moderately differentiated and 30 poorly differentiated. The expression of CREB and CXCR2 was evaluated by immunohistochemical Elivision method in lung squamous cell carcinoma and adenocarcinoma tissue (n=49)and in benign lung lesion tissue (n=20).We analyzed the expression and correlation between the groups according to the positive rate and intensity of lung cancer tissue.Results CREB positive expression was located mainly in cytoplasm and nuclei,CXCR2 mainly in cytoplasm.The expression of CREB and CXCR2 in lung cancer was significantly higher than that in benign lung lesions (P<0.01).The expression of CREB and CXCR2 in different degrees of differentia-tion of lung cancer had significant difference (P<0.01),and the expression of CREB and CXCR2 in poorly differentiated lung cancer tissue was significantly higher than that in the well-moderately differentiated (P<0.05).Conclusion CXCR2 and CREB are highly expressed in lung squamous cell carcinoma and adeno-carcinoma.With the reduction of lung tissue differentiation degree,the expressions of CREB and CXCR2 was significantly enhanced.
