目的:观察神经生长因子(NGF)及血管内皮生长因子(VEGF)在糖尿病大鼠下颌下腺内表达变化,探讨胰岛素对其表达影响及可能机制。方法 SD大鼠30只,随机分为:对照组、糖尿病组、胰岛素治疗组。大鼠2个月成模后,取血检测血糖;取下颌下腺组织,分别进行 HE染色、免疫组织化学染色和计算机图像分析。结果①对照组血糖与糖尿病组比较差异有统计学意义;胰岛素治疗组血糖与糖尿病组比较差异有统计学意义。②糖尿病组腺泡轻度萎缩,排列不规则,颗粒曲管数目减少,直径变小;胰岛素治疗组较对照组变化不明显。③与对照组相比较,糖尿病组NGF表达下调,VEGF表达上调。与糖尿病组相比较,胰岛素治疗组NGF表达上调,VEGF表达下调。结论糖尿病大鼠下颌下腺内NGF表达减少、VEGF表达增高,可能是糖尿病患者神经病变与血管病变的重要原因之一;胰岛素治疗可能与 NGF和 VEGF的表达改变有内在联系。

목적:관찰신경생장인자(NGF)급혈관내피생장인자(VEGF)재당뇨병대서하합하선내표체변화,탐토이도소대기표체영향급가능궤제。방법 SD대서30지,수궤분위:대조조、당뇨병조、이도소치료조。대서2개월성모후,취혈검측혈당;취하합하선조직,분별진행 HE염색、면역조직화학염색화계산궤도상분석。결과①대조조혈당여당뇨병조비교차이유통계학의의;이도소치료조혈당여당뇨병조비교차이유통계학의의。②당뇨병조선포경도위축,배렬불규칙,과립곡관수목감소,직경변소;이도소치료조교대조조변화불명현。③여대조조상비교,당뇨병조NGF표체하조,VEGF표체상조。여당뇨병조상비교,이도소치료조NGF표체상조,VEGF표체하조。결론당뇨병대서하합하선내NGF표체감소、VEGF표체증고,가능시당뇨병환자신경병변여혈관병변적중요원인지일;이도소치료가능여 NGF화 VEGF적표체개변유내재련계。
Obj ective To investigate the expression and significance of NGF and VEGF in the sub-mandibular glands of diabetic rats,and to study the effects of insulin on the expression and the possible mechanism.Methods 30 Sprague-Dawley rats were randomly divided into group C,group DM and group DM-I.After two months,all rats were killed for assaying blood sugar,then the submandibular glands were taken for observing the results using HE staining,immunohistochemical staining and image analysis by computer.Results ①The difference in the blood glucose between group C and group DM was significant (P<0.05),and the difference between group DM and group DM-I was significant (P<0.05).②The aci-nus mildly atrophied,and the acinar cells were arranged irregularly.The number and diameter of the tu-bules was reduced in group DM.But group DM-I was very close to group C.③Compared with those of group C,the expression of NGF was down-regulated and VEGF up-regulated in group DM.Compared with those of group DM,the expression of NGF was up-regulated and VEGF down-regulated in group DM-I. Conclusion The expression of NGF was increased and VEGF decreased in the submandibular glands of hy-perlipidemic rats,which may be one of the important reasons for neuropathy and vasculopathy.Insulin may have internal relations with the expression change of NGF and VEGF.
