目的:研究颅内应用神经生长因子(nerve growth factor,NGF)对大鼠脑出血(intracerebral hemorrhage)后血肿灶周围神经元的保护作用,并探讨不同剂量之间的差别。方法雄性SD大鼠,采自体血(50μl)注入右侧尾状核制作脑出血模型。根据不同注射剂量随机分为4组,生理盐水组(Control组)、0.5ug-NGF组、1.0ug-NGF组、1.5ug-NGF组。各组分别通过颅内原位注射等体积(2ul)生理盐水、NGF 0.5ug、NGF 1.0ug、NGF 1.5ug。治疗一周后进行神经功能评分(NDS);采用免疫组织化学技术观察血肿周NEURON阳性细胞的表达。结果①NDS评分:颅内注射不同剂量NGF组与单纯注射生理盐水的脑出血组相比,可明显改善神经功能缺损评分,差异有统计学意义(P<0.05),其中,以1.0ug-NGF组及1.5ug-NGF组最为明显,而这两者之间相比无统计学差异。②Neuron阳性细胞数:颅内注射不同剂量NGF组与单纯注射生理盐水的脑出血组相比,血肿灶周围的Neuron阳性细胞数明显增多,差异有统计学意义(P<0.05),其中,以1.0ug-NGF组及1.5ug-NGF组增加明显,而这两者之间相比无统计学差异。结论①颅内注射NGF能够改善脑出血后神经功能缺损及血肿灶周围神经元的损伤;②高剂量NGF治疗效果优于较低剂量。

목적:연구로내응용신경생장인자(nerve growth factor,NGF)대대서뇌출혈(intracerebral hemorrhage)후혈종조주위신경원적보호작용,병탐토불동제량지간적차별。방법웅성SD대서,채자체혈(50μl)주입우측미상핵제작뇌출혈모형。근거불동주사제량수궤분위4조,생리염수조(Control조)、0.5ug-NGF조、1.0ug-NGF조、1.5ug-NGF조。각조분별통과로내원위주사등체적(2ul)생리염수、NGF 0.5ug、NGF 1.0ug、NGF 1.5ug。치료일주후진행신경공능평분(NDS);채용면역조직화학기술관찰혈종주NEURON양성세포적표체。결과①NDS평분:로내주사불동제량NGF조여단순주사생리염수적뇌출혈조상비,가명현개선신경공능결손평분,차이유통계학의의(P<0.05),기중,이1.0ug-NGF조급1.5ug-NGF조최위명현,이저량자지간상비무통계학차이。②Neuron양성세포수:로내주사불동제량NGF조여단순주사생리염수적뇌출혈조상비,혈종조주위적Neuron양성세포수명현증다,차이유통계학의의(P<0.05),기중,이1.0ug-NGF조급1.5ug-NGF조증가명현,이저량자지간상비무통계학차이。결론①로내주사NGF능구개선뇌출혈후신경공능결손급혈종조주위신경원적손상;②고제량NGF치료효과우우교저제량。
Objective Investigate the protective effect of Intracerebral injecting Nerve Growth Factor (NGF) on Intracerebral Hemorrhage of rats, and discuss the differences among injecting in different dose. Methods The cerebral hemorrhage (ICH) model was induced by stereotaxic infusion of 50μl autologous blood into the right striatum. Divide them into 4 groups according to differenting dose, namely control group, 0.5ug-NGF group, 1.0ug-NGF group, 1.5ug-NGF group. Each group is injected the same dose (2ul) of normal saline, ngf 0.5ug, ngf 1.0ug, ngf 1.5ug by Intracerebral injection. Neurological Deficit Scale(NDS), immunohistochemical technique were used to detect the expression of NEURON, 1 week after administration respectively. Results ①NDS score: Groups of Intracerebral injecting different dose ngf have apparent decrease with purely injecting normal saline group in NDS evaluation, and the difference is statistical y significance(p<0.05), especial y that of the 1.0ug and 1.5ug groups, but the two groups have no statistical y significance. ②Neuron positive cel s: Groups of Intracerebral injecting different dose ngf have apparent improvement with purely injecting normal saline group in the number of Neuron positive cel s in perihematoma region. and the improvement is statistical y significance(p<0.05), especial y that of the 1.0ug and 1.5ug groups, but the two groups have no statistical y significance. Conclusion: ①Intracerebral injecting of ngf can treat the defect of neurological function and neuron damage around perihematoma region after intracerebral hemorrhage. ②High dose of ngf has better treatment effect than lower dose.
