采用Nishikawa法,从成都郊区土壤中筛选获得一株ε-聚赖氨酸(ε-PL)产量为1.893g/L的菌株SAC23,通过16S rRNA分子鉴定,初步确定该菌株为Streptomyces ahygroscopicus (不吸水链霉菌)。以SAC23为出发菌株,通过60Coγ射线诱变,结合亚甲基蓝与链霉素抗性平板筛选,获得1株遗传性能稳定,ε-PL产量可达4.384g/L的变异菌株SAS17,与SAC23相比较,ε-PL产量提高了131.6%。表明通过60Coγ射线诱变,结合亚甲基蓝与链霉素抗性平板筛选可大幅提高链霉菌生产ε-聚赖氨酸的能力。

A ε-polylysine (ε-PL) producer strain SAC23 with 1.893g/L yield, was screened from the soil collected from the suburbs of Chengdu, by using Nishikawa's method. The phylogenetic tree based on the 16S rRNA se-quences indicates that it belongs to Streptomyces ahygroscopic. A mutant SAS17 with high-production of ε-PL and stable genetic performance was screened by 60Co ray combining drug-resistance technique and methylene blue plate. The production of ε-PL reached to 4.384g/L, increasing 131.6% compared with that of the original strain SAC23. It shows that the production of ε-polylysine of ahygroscopicus can be improved significantly through the method of 60Co mutagenesis combined with streptomycin resistance plate.
