目的:探讨齐墩果酸及环磷酰胺对H-22荷瘤小鼠的抗肿瘤作用机制。方法:肝癌H-22肿瘤细胞的小鼠随机分为模型组、环磷酰胺组、齐墩果酸组、齐墩果酸组与环磷酰胺联合应用组。连续给药8天后剥离瘤组织,采用免疫组化法检测肿瘤组织中突变型P53蛋白及肿瘤坏死因子( TNF-α)的含量。结果:齐墩果酸组与环磷酰胺联合应用对H-22荷瘤小鼠TNF-α含量明显提高,与模型组比较差异显著( P<0.05);齐墩果酸组与环磷酰胺联合应用对H-22荷瘤小鼠突变型P53蛋白表达明显降低,与模型组比较差异显著(P<0.05)。结论:齐墩果酸组与环磷酰胺联合应用能下调H-22荷瘤小鼠肿瘤细胞内突变型P53蛋白的表达,对H-22荷瘤小鼠肿瘤细胞内TNF-α有明显的诱生作用。

목적:탐토제돈과산급배린선알대H-22하류소서적항종류작용궤제。방법:간암H-22종류세포적소서수궤분위모형조、배린선알조、제돈과산조、제돈과산조여배린선알연합응용조。련속급약8천후박리류조직,채용면역조화법검측종류조직중돌변형P53단백급종류배사인자( TNF-α)적함량。결과:제돈과산조여배린선알연합응용대H-22하류소서TNF-α함량명현제고,여모형조비교차이현저( P<0.05);제돈과산조여배린선알연합응용대H-22하류소서돌변형P53단백표체명현강저,여모형조비교차이현저(P<0.05)。결론:제돈과산조여배린선알연합응용능하조H-22하류소서종류세포내돌변형P53단백적표체,대H-22하류소서종류세포내TNF-α유명현적유생작용。
Objective:To investigate the antitumor mechanism of oleanolic acid and cyclophosphamide on H -22 tumor bearing mice .Methods:H-22 tumor cells were randomly divided into model group , cyclophosphamide group , oleanolic acid group , combined group of oleanolic acid and cyclophosphamide .Continuous administration for eight days after the tumor tissue , immunohistochemistry of mutant P 53 protein and tumor necrosis factor in the tumor tissue were detected ( TNF-α) content .Results:By combined application of oleanolic acid and cyclophosphamide in mice H -22 TNF-αcontent increased significantly , and compared with the model group it had a significant difference (P<0.05);by com-bined application of oleanolic acid and cyclophosphamide the expression of H -22 mutant P53 protein was significantly decreased in mice, with marked difference compared with the model group (P<0.05).Conclusion:The expression of combined application of oleanolic acid and cyclophosphamide can down -regulate H-22 in tumor cells of mutant P 53 protein,which has an effect on the H -22 induced tumor cell of tumor bearing mice in TNF -α.
