为探明烟草内生细菌种群特征及其与烟草的关系,采用研磨组织稀释分离法分离不同品种、不同生长期、不同器官的烟草内生细菌,以各菌群出现与否为指标,用类平均法进行系统聚类分析,对内生细菌的种类组成,分布特征进行了研究。结果表明,烟草内生细菌种类组成存在品种差异、生长期差异和组织部位差异;优势菌群随分离部位和生长时期的不同而变化。Bacillus sp.和Rhizobiumsp.是4个品种烟草叶片共有的优势类群,从苗期到团棵期,内生细菌的种类在增加,B. pumilus广泛存在于成熟期烟草的根、茎、叶中,且为主要菌群。部分内生细菌具有组织偏好性,Micrococcus sp.和Staphylococcus sp.对烟草的茎部具有偏好性,B. cereus对烟草叶片具有偏好性。研究结果为烟草有益内生细菌的筛选奠定了基础,并为内生细菌在其它植物中的开发利用提供理论依据。

Endophytic bacteria were isolated from tobacco by the method of grinding separation. The composition of endophytic bacteria population in different cultivars, different tissues and different growth stages were studied. Results showed that the composition and quantity of dominant bacterial community varied with changes of cultivars, growth stages and tissues.Bacillus sp. andRhizobium sp. were considered as dominant genera of endophytic bacteria in leaf from different tobacco cultivars. The endophytic bacteria flora in leaf increased from seedling stage to maturing stage.B. pumilus were considered as dominant genera of endophytic bacteria in root, stem and leaf during maturing stage. Cluster analysis in which euclidean distance was taken as similarity measure using group average method showed that endophytic bacteria in tobacco had tissue specificity.Micrococcus sp. andStaphylococcus sp. had tissue specificity to stem whileBacillus cereus had tissue specificity to leaf. The study could help to lay foundation for screening beneficial endophytic bacteria and to provide references for using in other crop plants.
