目的:建立测定人血清中苯妥英钠浓度的方法。方法:采用大连依利特HypersilODS2色谱柱(4.6mm×150mm,5μm ),流动相为甲醇-乙腈-水(20∶15∶65),流速为1mL·min-1,检测波长为210nm,柱温为30℃,内标为苯巴比妥。结果:苯妥英钠血药浓度在6.25~100 mg·L-1范围内线性关系良好(r=0.9995,n=6),最低检测浓度为1.56mg·L-1;平均回收率为99.64%~100.2%,日内与日间RSD均<5%(n=5)。结论:本方法快速、简便、准确,适用于临床苯妥英钠的血药浓度监测。

Objective:To establish the method for the concentration determination of phenytoin sodium in human serum. Methods: The chromatographic column was Hypersil ODS2 (150 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-acetonitrile-water (20:15:65) at flow rate of 1.0 mL?min-1. The colum temperature was 30℃ and the detection wavelength was set at 210 nm. The phenobarbit was used as internal standard. Results: Blood concentrations of phenytoin was within the detection ranges of 6.25 ~ 100 mg?L-1(r=0.999 5,n=6).The ratios of average recovery was 99.64% ~ 100.2% and precision within a day and among days were less than 5.0%.Conclusion: The method is rapid, simple, accurate and suitable for routine monitoring of clinical need.
