目的:建立超高效液相色谱法测定全血中环孢素A的质量浓度。方法血样经乙醚萃取后,经WATERS BEH C18(2.1×50mm,1.7μm)分离,乙腈-水为流动相,70℃柱温下进行色谱分析,检测波长为210nm。结果本方法线性范围为0.05~1.0μg· mL -1(r=0.9930),高、中、低血样的平均回收率分别为94.04%、100.81%、98.95%,日内RSD小于10%。结论本法操作简便,回收率高,精密度好,适用于临床常规检测CsA的血药浓度。

목적:건립초고효액상색보법측정전혈중배포소A적질량농도。방법혈양경을미췌취후,경WATERS BEH C18(2.1×50mm,1.7μm)분리,을정-수위류동상,70℃주온하진행색보분석,검측파장위210nm。결과본방법선성범위위0.05~1.0μg· mL -1(r=0.9930),고、중、저혈양적평균회수솔분별위94.04%、100.81%、98.95%,일내RSD소우10%。결론본법조작간편,회수솔고,정밀도호,괄용우림상상규검측CsA적혈약농도。
OBJECTIVE To establish a UPLC method for detecting cyclosporine A concentration in human whole blood.METHODS CsA was extracted from whole blood with diethyl and separated by a WATERS BEH C 18 (2.1 ×50mm,1.7μm).Mobile phase was a mixture of acetonitrile and water ,the column temperature was set at 70℃,and the detected wavelength was at 210 nm.RESULTS The average linear range of CsA was from 0.05μg· mL-1 to 1.0μg · mL-1 , r =0.9930.The recovery rate of low、moderate、high concentrations were 94.04%、100.81%、98.95% respectively.The intraday RSD were 5.0%( less than 10%) ( n=6 ).CONCLUSION This method is simple ,accurate ,precise and suitable for monitoring blood concentration of CsA.
