目的：探讨高尔基体糖蛋白-73（GP73）和人第10号染色体缺失的磷酸酯酶及张力蛋白同源蛋白（PTEN）在原发性肝细胞癌（HCC）患者血清中的表达及其临床意义，并分析甲胎蛋白（AFP）、GP73和PTEN 三者联合检测在 HCC 早期诊断中的意义。方法用ELISA法检测 HCC组（n＝90）、肝硬化组（n＝99）、肝炎组（n＝90）和健康对照组（n＝81）的血清GP73和PTEN浓度，用微粒子免疫化学发光法检测各组血清AFP浓度。结果 HCC组GP73和AFP浓度均高于其他3组，PTEN浓度低于其他3组，差异均有统计学意义（P＜0．01）。HCC 患者血清 GP73和 AFP 的浓度与肿瘤大小呈正相关（rGP73＝0．343，PGP73＝0．002；rAFP＝0．250，PAFP＝0．034）。GP73和 PTEN 二者联合检测的灵敏度、特异度和符合率分别为94．44％、65．93％和73．06％，优于其他组合。结论 GP73和PTEN是较敏感特异的良好 HCC标志物，两者联合检测可提高 HCC诊断的敏感性。
목적：탐토고이기체당단백-73（GP73）화인제10호염색체결실적린산지매급장력단백동원단백（PTEN）재원발성간세포암（HCC）환자혈청중적표체급기림상의의，병분석갑태단백（AFP）、GP73화PTEN 삼자연합검측재 HCC 조기진단중적의의。방법용ELISA법검측 HCC조（n＝90）、간경화조（n＝99）、간염조（n＝90）화건강대조조（n＝81）적혈청GP73화PTEN농도，용미입자면역화학발광법검측각조혈청AFP농도。결과 HCC조GP73화AFP농도균고우기타3조，PTEN농도저우기타3조，차이균유통계학의의（P＜0．01）。HCC 환자혈청 GP73화 AFP 적농도여종류대소정정상관（rGP73＝0．343，PGP73＝0．002；rAFP＝0．250，PAFP＝0．034）。GP73화 PTEN 이자연합검측적령민도、특이도화부합솔분별위94．44％、65．93％화73．06％，우우기타조합。결론 GP73화PTEN시교민감특이적량호 HCC표지물，량자연합검측가제고 HCC진단적민감성。
Objective To investigate the clinical significance of serum Glogi glycoprotein 73 (GP73)and phosphatase and tensin homolog deleted in chromosome 10 (PTEN)for the diagnosis of hepatocellular carcinoma(HCC).Methods Peripheralserum sam-ples,collected from 90 HCC patients,99 cirrhotic patients,90 hepatitis patients and 81 healthy subjects,were detected for GP73 and PTEN levels by using enzyme-linked immunosorbent assays and for alpha fetoprotein (AFP)levels by using microparticle chemilu-minescent assay.Results Serum levels of GP73 and AFP in HCC patients were significantly higher than the other three groups,but PTEN level was lower (P<0.01).Serum levels of GP73 and AFP were positively correlated with the tumor size (rGP73= 0.343, PGP73= 0.002;rAFP= 0.250,PAFP= 0.034).Sensitivity,specificity,and coincidence rate of combined detection of GP73 and PTEN were 94.44%,65.93% and 73.06%,which were better than any other combinations.Conclusion GP73 and PTEN could be sensi-tive and specific markers in the diagnosis of HCC,and combined examination of GP73 and PTEN could improve the sensitivity for the diagnosis of HCC.
