Bailing capsule%IgA nephropathy rats%Immunologic derangement
目的:通过建立IgA肾病大鼠模型,探讨百令胶囊对改善IgA大鼠肾功能水平及免疫紊乱状况的效果。方法:选取成年雄性健康的SD大鼠20只为研究对象,通过皮下注射四氯化碳(CCL4)、口服牛血清白蛋白(BSA)以及静脉注射脂多糖(LSP)复合法建立IgA肾病大鼠模型。将大鼠分为健康对照组(NC组)、泼尼松治疗组(PN组)、百令胶囊治疗组(BN组)以及模型对照组(NS组),每组5只。PN组及BN组于胃部灌注泼尼松及百令胶囊,NC组及NS组于胃部灌注同等剂量的蒸馏水。观察各组大鼠治疗前后肾功能指标及血脂水平的变化。采用ELISA酶联测定各组大鼠实验前后Th1类细胞因子IL-2、INF-γ以及Th2类细胞因子IL-4、IL-10水平。结果:BN组治疗后血SCr、血BUN、尿RBC水平低于PN组及NS组,差异有统计学意义(P<0.05),而与NC组相比差异无统计学意义(P>0.05)。治疗后BN组TC、TG、LDL水平显著低于PN组及NS组,而HDL则高于PN组及NS组,差异有统计学意义(P<0.05),与对照组比较差异无统计学意义(P>0.05)。BN组IL-2、INF-γ水平显著高于PN组及NS组,而IL-4、IL-10显著低于PN组及NS组,差异有统计学意义(P<0.05)。结论:百令胶囊能有效降低IgA肾病大鼠尿蛋白及血脂水平,改善患者肾脏功能,并可调节IgA肾病大鼠免疫系统功能,改善大鼠炎性因子水平,从而起到治疗IgA肾病的效果。

목적:통과건립IgA신병대서모형,탐토백령효낭대개선IgA대서신공능수평급면역문란상황적효과。방법:선취성년웅성건강적SD대서20지위연구대상,통과피하주사사록화탄(CCL4)、구복우혈청백단백(BSA)이급정맥주사지다당(LSP)복합법건립IgA신병대서모형。장대서분위건강대조조(NC조)、발니송치료조(PN조)、백령효낭치료조(BN조)이급모형대조조(NS조),매조5지。PN조급BN조우위부관주발니송급백령효낭,NC조급NS조우위부관주동등제량적증류수。관찰각조대서치료전후신공능지표급혈지수평적변화。채용ELISA매련측정각조대서실험전후Th1류세포인자IL-2、INF-γ이급Th2류세포인자IL-4、IL-10수평。결과:BN조치료후혈SCr、혈BUN、뇨RBC수평저우PN조급NS조,차이유통계학의의(P<0.05),이여NC조상비차이무통계학의의(P>0.05)。치료후BN조TC、TG、LDL수평현저저우PN조급NS조,이HDL칙고우PN조급NS조,차이유통계학의의(P<0.05),여대조조비교차이무통계학의의(P>0.05)。BN조IL-2、INF-γ수평현저고우PN조급NS조,이IL-4、IL-10현저저우PN조급NS조,차이유통계학의의(P<0.05)。결론:백령효낭능유효강저IgA신병대서뇨단백급혈지수평,개선환자신장공능,병가조절IgA신병대서면역계통공능,개선대서염성인자수평,종이기도치료IgA신병적효과。
Objective:To explore the effect of Bailing capsule in the improvement of kidney function and immunologic derangement of IgA rats through the establishment of IgA nephropathy rat model.Methods:20 cases of adult male health SD rats were selected as the research objects.IgA nephropathy rat model was established by subcutaneous injection of carbon tetrachloride(CCL4),oral administration of bovine serum albumin(BSA) and intravenous injection of lipopolysaccharide(LSP).The rats were divided into the normal control group(NC group),the prednisone treatment group(PN group),the Bailing capsule treatment group(BN group) and the model control group(NS group) with 5 rats in each.PN group and BN group were given stomach perfusion of prednisone and Bailing capsule.NC group and NS group were given stomach perfusion of the same dose of distilled water.The changs of renal function indices and blood lipid levels before and after treatment were observed in the rats.The Th1 cytokines IL-2,INF-γ and Th2 cytokines IL-4,IL-10 levels before and after experiment were measured by ELISA enzymeimmunoassay in each group of rats.Results:After treatment,the blood SCr,blood BUN,urine RBC levels of BN group were lower than those of PN group and NS group,and the difference was statistically significant.But there had no statistical significance compared with NC group(P>0.05).After treatment,TC,TG,LDL levels of BN group were significantly lower than those of PN group and NS group,but HDL was higher than that of PN group and NS group,and the difference was statistically significant(P<0.05), and there had no statistical significance compared with the control group(P>0.05).IL-2,INF-γ levels of BN group were significantly higher than those of PN group and NS group,but IL-4,IL-10 levels were significantly lower than those of PN group and NS group,and the difference was statistically significant(P<0.05).Conclusion:Bailing capsule can effectively reduce urinary protein and lipid levels of IgA nephropathy rats,and improve renal function of patients.It may regulate immune system function of IgA nephropathy rats,and improve inflammatory factor levels of rats,thereby play the treatment effect of IgA nephropathy.
