以脱壳蓖麻籽为原料,在单因素实验的基础上,采用响应面法优化蓖麻粗毒蛋白的提取工艺。选取液料比、PH、浸提时间和硫酸铵饱和度4因素3水平进行中心组合实验。结果表明,蓖麻粗毒蛋白的最佳提取工艺条件为:液料比4.5 mL/g,PH 6.9,浸提时间38 h,硫酸铵饱和度65%。优化后蛋白提取率从2.82%提高到3.69%, SDS-PAGE凝胶电泳显示32 kD和34 kD两条蛋白带。

Based on the single factor exPeriments,the resPonse surface methodology was emPloyed to oPti-mize the extraction Process for ricin from caster beans. The ratio of solid-to-solvent,PH,extraction time and ammonium sulfate saturation were used to carry out a 4-variable,3-level central comPosite design. The result showed that the oPtimal conditions were ratio of solid-to-solvent 4. 5 mL/g,PH 6. 9,extraction time 38 h and ammonium sulfate saturation 65%. Protein extraction yield under the oPtimal conditions was im-Proved from 2 . 82% to 3 . 69%. SDS-PAGE analysis showed that raw ricin contained 32 kD and 34 kD Protein band.
