Alfalfa polysaccharides%Immunization%Effect
为了研究苜蓿多糖对新城疫弱毒苗免疫鸡所产生的免疫调节作用,本研究选择1日龄海兰雏鸡120羽,随机分为4组,每组30羽,7日龄每羽用鸡新城疫活疫苗(La Sota株)免疫,28日龄以相同剂量进行加强免疫。其中3个试验组从7日龄开始分别肌肉注射低、中、高剂量(10、25、50μg/mL)的苜蓿多糖,对照组注射生理盐水,1次/d,连用7 d。于14、21、28、35、42、49、56日龄,通过静脉采血,用MTT法检测外周血淋巴细胞吸光度值(OD值);同时用微量法检测血清中新城疫病毒血凝抑制(HI)抗体效价。结果显示,苜蓿多糖能够显著提高外周血淋巴细胞OD值,中剂量组(25μg/mL)淋巴细胞OD值最大;苜蓿多糖能够显著提高血清中新城疫病毒HI抗体效价,中剂量组免疫后期抗体存在时间长,抗体高峰期持续时间也长。本试验表明合适剂量苜蓿多糖对新城疫弱毒苗的免疫效果有明显的增强作用。

위료연구목숙다당대신성역약독묘면역계소산생적면역조절작용,본연구선택1일령해란추계120우,수궤분위4조,매조30우,7일령매우용계신성역활역묘(La Sota주)면역,28일령이상동제량진행가강면역。기중3개시험조종7일령개시분별기육주사저、중、고제량(10、25、50μg/mL)적목숙다당,대조조주사생리염수,1차/d,련용7 d。우14、21、28、35、42、49、56일령,통과정맥채혈,용MTT법검측외주혈림파세포흡광도치(OD치);동시용미량법검측혈청중신성역병독혈응억제(HI)항체효개。결과현시,목숙다당능구현저제고외주혈림파세포OD치,중제량조(25μg/mL)림파세포OD치최대;목숙다당능구현저제고혈청중신성역병독HI항체효개,중제량조면역후기항체존재시간장,항체고봉기지속시간야장。본시험표명합괄제량목숙다당대신성역약독묘적면역효과유명현적증강작용。
In order to observe the immunoregulatory effects of alfalfa polysaccharide on Newcas-tle disease vaccine immunization, 120 1-day-age chickens were selected and randomly divided into four groups, 30 chickens per group. La Sota strain immunization and booster were performed at day-7and day-28. From day-7, three test group chickens were inoculated diffraction dose (10, 25, 50 μg/mL) of alfalfa polysaccharide muscle injection each day, and control chickens were inject-ed physiological saline water synchronously.At the day-14, 21, 28, 35, 42, 49 and 56, all chick-ens were blooded and the peripheral blood lymphocyte absorbance value (OD value) was detected by MTT method. The serum antibody titers of NDV were determined by hemagglutination inhibition (HI) method. As a results, alfalfa polysaccharide can improve the OD value of peripheral blood lympho-cytes transformation and the OD values most significantly with dose of 25 μg/mL, meanwhile, al-falfa polysaccharide can significantly increase the HI antibody titer in serum and maintain late high immune antibody level long time, antibody peak period showed longer than control groups. The experiment showed that appropriate doses of alfalfa polysaccharides obviously enhanced the immune effect of Newcastle disease vaccine immunization effect.
