目的:探讨加味导赤散与维生素B12联合治疗复发性口腔溃疡(ROU)动物模型的作用及其对血清IL-6及TNF-α的影响。方法:采用免疫方法建立ROU动物模型。48只SD大鼠随机分为正常组、模型组、左旋咪唑组(对照组)、低剂量加味导赤散加维生素B12组(治疗组1)、中剂量加味导赤散加维生素B12组(治疗组2)、高剂量加味导赤散加维生素B12组(治疗组3),每组8只。对照组用左旋咪唑片灌胃,治疗组分别用低、中、高剂量的加味导赤散与维生素B12一起灌胃,观察记录溃疡变化情况,用酶联免疫法(ELISA)测定大鼠造模前后及灌胃后血清IL-6及TNF-α含量。结果:①.实验大鼠于第3次注射抗原乳化剂1周后开始出现口腔溃疡。②.对照组与治疗组2、组3的大鼠口腔溃疡分别较模型组发生数目明显减少,持续时间明显缩短,间隔时间明显延长(P<0.05)。③.模型组IL-6及TNF-α水平较正常组明显升高(P<0.05),而对照组与2、3治疗组较溃疡组下降(P<0.05)。结论:加味导赤散加维生素B12可能通过下调IL-6及TNF-α水平促进口腔溃疡愈合,减少溃疡复发。

목적:탐토가미도적산여유생소B12연합치료복발성구강궤양(ROU)동물모형적작용급기대혈청IL-6급TNF-α적영향。방법:채용면역방법건립ROU동물모형。48지SD대서수궤분위정상조、모형조、좌선미서조(대조조)、저제량가미도적산가유생소B12조(치료조1)、중제량가미도적산가유생소B12조(치료조2)、고제량가미도적산가유생소B12조(치료조3),매조8지。대조조용좌선미서편관위,치료조분별용저、중、고제량적가미도적산여유생소B12일기관위,관찰기록궤양변화정황,용매련면역법(ELISA)측정대서조모전후급관위후혈청IL-6급TNF-α함량。결과:①.실험대서우제3차주사항원유화제1주후개시출현구강궤양。②.대조조여치료조2、조3적대서구강궤양분별교모형조발생수목명현감소,지속시간명현축단,간격시간명현연장(P<0.05)。③.모형조IL-6급TNF-α수평교정상조명현승고(P<0.05),이대조조여2、3치료조교궤양조하강(P<0.05)。결론:가미도적산가유생소B12가능통과하조IL-6급TNF-α수평촉진구강궤양유합,감소궤양복발。
Objective:①.Establish the rat's recurrent oral ulceration(ROU) models;②.To evaluate the therapeutic ef-fects of traditional chinese medicine Dao chi San plus gypsum decoction (DCSGd) with vitamin B12 on experimental oral ul-cer. Method:Experimental animal model of ROU was established by immunological method (by injecting antigens into SD rats). 48 rats were randomized into six groups,including normal group,ulcer group (model group), Levamisole group,low-dose DCSGd plus vitamin B12 group (therapy group 1),medium-dose DCSGd plus vitamin B12 group (therapy group 2),and high-dose DCSGd plus vitamin B12 group(therapy group 3). All groups were taken orally. During these days,observe general symptoms and the oral ulcers change of rats in each group. The therapeutic effects were observed and the level of IL-6 and TNF-αwere measured in different groups. Result:①.the ulcers were formed one week after the third injection.②.Compared with ulcer group,the number of ulcers are reduced,interval are increased and the duration are shorten in Levamisole group, therapy group 2 and therapy group 3. ③.Compared with normal group,the expression intensity of IL-6 and TNF-αin model group was significantly higher(P<0.05). The level of IL-6 and TNF-αwere decreased in Levamisole group,therapy group 2 and therapy group 3,compared with model group(P<0.05). Conclusion:The abnormal increase of IL-6 and TNF-αis relat-ed to ROU. DCSGd plus vitamin B12 can promote ulcer healing, reduce symptoms and reduce ulcer recurrence.
