目的:探索丁苯酞对弥漫性脑损伤大鼠模型脑水肿及皮层基质金属蛋白酶9( matrix metalloproteinase-9,MMP-9)、咬合蛋白表达的影响。方法 Sprague-Dawley 雄性大鼠140只,随机分为对照组和丁苯酞组(40mg/kg,1次/d)各70只。应用Marmarou法建立弥漫性脑损伤大鼠模型。2组分别于术后6、12、24、72、144h采用干湿质量法检测大鼠脑组织含水量变化,采用免疫组织化学检测损伤区周围皮层MMP-9和咬合蛋白表达变化。2组分别于术后6、24、72、144h采用Western blot法检测损伤区周围皮层MMP-9和咬合蛋白含量变化。结果与对照组比较,丁苯酞组脑组织含水量降低,损伤区周围皮层MMP-9表达减少( P﹤0.05),咬合蛋白表达增多( P﹤0.05)。结论丁苯酞可降低弥漫性脑损伤大鼠皮层MMP-9的表达,增加咬合蛋白的表达,减轻脑水肿的形成。

목적:탐색정분태대미만성뇌손상대서모형뇌수종급피층기질금속단백매9( matrix metalloproteinase-9,MMP-9)、교합단백표체적영향。방법 Sprague-Dawley 웅성대서140지,수궤분위대조조화정분태조(40mg/kg,1차/d)각70지。응용Marmarou법건립미만성뇌손상대서모형。2조분별우술후6、12、24、72、144h채용간습질량법검측대서뇌조직함수량변화,채용면역조직화학검측손상구주위피층MMP-9화교합단백표체변화。2조분별우술후6、24、72、144h채용Western blot법검측손상구주위피층MMP-9화교합단백함량변화。결과여대조조비교,정분태조뇌조직함수량강저,손상구주위피층MMP-9표체감소( P﹤0.05),교합단백표체증다( P﹤0.05)。결론정분태가강저미만성뇌손상대서피층MMP-9적표체,증가교합단백적표체,감경뇌수종적형성。
Objective To explore the effect of butylphthalide on the brain edema of rats following traumatic brain injury and the expression of cortical matrix metalloproteinase-9( MMP-9 )and occludin around the injury area. Methods One hundred and forty Sprague-Dawley male rats were divided into control group and butylphthalide group(40mg/kg,once a day). The model was made by Marmarou method. After injury 6h,12h,24h,72h and 144h,brain water content was measured to observe the changes of the brain edema,the immunohistochemistry and Western blot were used to observe the expression of MMP-9 and occludin around the injury area. Results Compared with the control group,the brain water content of the butylphthalide group was less,the content of MMP-9 around the injury area decreased significantly( P﹤0. 05 ). And the content of occludin around the injury area increased( P﹤0. 05 ). Conclusion Butylphthalide could reduce the expression of MMP-9 following traumatic brain injury,increase the expression of MMP-9,and reduce the brain edema.
