目的:观察穗花杉双黄酮对人结肠癌细胞SW480增殖及凋亡相关因子caspase-3、β-catenin的影响,初步探讨其诱导结肠癌细胞凋亡的作用机制。方法 MTT细胞增殖试验检测穗花杉双黄酮对SW480细胞活力的影响,流式细胞术检测其对SW480细胞凋亡的诱导作用,Western blot法检测穗花杉双黄酮作用细胞后caspase-3、β-catenin蛋白的表达。结果穗花杉双黄酮对SW480细胞生长有抑制作用,具有剂量依赖性。150μmol/L穗花杉双黄酮明显促进其凋亡,凋亡率显著高于对照组(P<0.01)。Western bolt结果表明,穗花杉双黄酮干预可以使caspase-3表达明显增强,β-catenin表达明显减弱。结论穗花杉双黄酮可诱导人结肠癌细胞SW480凋亡,并影响凋亡相关因子caspase-3、β-catenin的表达,从而达到抗肿瘤作用。

목적:관찰수화삼쌍황동대인결장암세포SW480증식급조망상관인자caspase-3、β-catenin적영향,초보탐토기유도결장암세포조망적작용궤제。방법 MTT세포증식시험검측수화삼쌍황동대SW480세포활력적영향,류식세포술검측기대SW480세포조망적유도작용,Western blot법검측수화삼쌍황동작용세포후caspase-3、β-catenin단백적표체。결과수화삼쌍황동대SW480세포생장유억제작용,구유제량의뢰성。150μmol/L수화삼쌍황동명현촉진기조망,조망솔현저고우대조조(P<0.01)。Western bolt결과표명,수화삼쌍황동간예가이사caspase-3표체명현증강,β-catenin표체명현감약。결론수화삼쌍황동가유도인결장암세포SW480조망,병영향조망상관인자caspase-3、β-catenin적표체,종이체도항종류작용。
Objective To investigate the role of β-catenin and caspase-3 in amentoflavone-induced apoptosis of human colorectal cancer SW480 cells. Methods MTT assay was used to detect the viability of SW480 cells exposed to amentoflavone, and flow cytometry was employed to assess the cell apoptosis. Western blotting was performed to determine the protein expressions ofβ-catenin and caspase-3 in the exposed cells. Results Amentoflavone dose-dependently inhibited the viability of SW480 cells, and a high concentration of amentoflavone (150μmol/L) obviously induced apoptosis of the cells. Amentoflavone exposure caused significantly increased expression of caspase-3 and suppressedβ-catenin expression in the cells. Conclusion Amentoflavone-induced apoptosis in SW480 human colorectal cancer cells is associated with altered expressions ofβ-catenin and caspase-3.
