为探索中华枸杞(Lycium chinense Mill. var. chinense)叶片离体再生技术,通过设置不同消毒时间梯度,研究了0.1%升汞溶液对中华枸杞叶片的消毒效果;以MS为基本培养基,添加6-BA、2,4-D、IAA等激素,研究了不同激素浓度配比对中华枸杞叶片离体再生的影响。结果表明,用0.1%升汞溶液消毒叶片8 min,污染率最低,是中华枸杞叶片的最佳消毒时间。基本培养基中添加1.0 mg/L 6-BA+0.2 mg/L 2,4-D是诱导愈伤组织的最佳激素浓度配比;基本培养基中添加1.0 mg/L 6-BA +0.1 mg/L NAA是诱导愈伤组织分化的最佳激素浓度配比;基本培养基中添加0.2 mg/L IAA +0.6 mg/L NAA是诱导丛生芽生根的适宜激素浓度配比。利用组织培养技术可有效实现中华枸杞叶片离体再生和快速繁殖,为其工厂化育苗和优良品种的选育奠定基础。

To explore the vitro regeneration from leaves of Lycium chinense Mill. var. chinense, a gradient of different disin-fection time have been set up to study the effect of the 0.1%mercuric chloride solution to disinfect the leaves of L. chinense Mill. var. chinense. Depending on the MS base medium, the plant hormones 6-BA,2,4-D and IAA were added in the MS. Through the orthogonal experiment, the effects of different plant hormones concentration ratio on the vitro regeneration from leaves of L. chinense Mill. var. chinense have been studied. The pollution rate was the lowest when the leaves were dealt with 0.1% mercuric chloride solution in 8 min (the best sterilization time). MS medium with 1.0 mg/L 6-BA and 0.2 mg/L 2,4-D was the best hormone concentration ratio to induce callus. MS medium with 1.0 mg/L 6-BA and 0.1 mg/L NAA was the best hormone concentration ratio to induce callus differentiation. Using tissue culture technology can not only effectively achieve the vitro regeneration and rapid propagation from leaves of L. chinense Mill. var. chinense, but also lay foundation for industrialized seedling production and the seed selection.
