目的:探讨津力达对胰岛素抵抗大鼠骨骼肌GLUT4表达的影响。方法 SD大鼠分为正常对照组( ND组),高脂对照组( HFD组),津力达组及吡格列酮组,每组6只;灌胃8周后,进行腹腔注射葡萄糖耐量试验并计算曲线下面积( AUC);测得大鼠空腹血糖、胰岛素计算HOMA-IR指数;RT-PCR及western-blotting检测大鼠骨骼肌GLUT4表达。结果津力达明显改善了30 min、60 min血糖及AUC,吡格列酮改善了60、120 min血糖及AUC( P <0.05);津力达及吡格列酮明显改善HOMA-IR指数( P <0.05);津力达及吡格列酮明显上调了骨骼肌GLUT4的表达。结论津力达能够上调大鼠骨骼肌GLUT4的表达,改善高脂饮食诱导的胰岛素抵抗。

목적:탐토진력체대이도소저항대서골격기GLUT4표체적영향。방법 SD대서분위정상대조조( ND조),고지대조조( HFD조),진력체조급필격렬동조,매조6지;관위8주후,진행복강주사포도당내량시험병계산곡선하면적( AUC);측득대서공복혈당、이도소계산HOMA-IR지수;RT-PCR급western-blotting검측대서골격기GLUT4표체。결과진력체명현개선료30 min、60 min혈당급AUC,필격렬동개선료60、120 min혈당급AUC( P <0.05);진력체급필격렬동명현개선HOMA-IR지수( P <0.05);진력체급필격렬동명현상조료골격기GLUT4적표체。결론진력체능구상조대서골격기GLUT4적표체,개선고지음식유도적이도소저항。
Objective To investigate the effect of Jinlida on the expression of GLUT 4 in skeletal muscle of rats with insulin resistance .Methods Sprague Dawley ( SD ) rats were randomly divided into four groups:normal diet group ( ND group),high-fat diet group (HFD group),high fat diet with Jinlida group (Jinlida group) and high fat diet with pioglitazone group (pioglitazone group),with 6 rats in each group.After 8 weeks,intraperitoneal glucose tolerance test (IPGTT) was performed and area under the curve ( AUC) was calculated based on the blood glucose curves;blood glucose and insulin were detected to calculate HOMA-IR;the expression levels of GLUT 4 in skeletal muscle were detected by quantitative real-time polymerase chain reaction (RT-PCR) and Western Blot.Results Jinlida administration obviously improved 30min,60min blood glucose and AUC during IPGTT,pioglitazone improved significantly 60min,120min blood glucose and AUC ( P <0.05),both Jinlida and pioglitazone improved obviously HOMA-IR index ( P <0.05),and both of them up-regulated the expression of GLUT4 in skeletal muscle ( P <0.05).Conclusion Jinlida can up-regulate expression of GLUT4 in skeletal muscle of rats,as a result,which can improve insulin resistance induced by high fat diet .
