目的:研究高效液相色谱法测定氯霉素醇溶液中氯霉素的含量。方法采用高效液相色谱法,以Eclipse XDB-C18(4.6 mm×250 mm,5μm)为色谱柱;流动相为0.01 mol/L庚烷磺酸钠缓冲液(取磷酸二氢钾6.8 g,用0.01 mol/L庚烷磺酸钠溶液溶解并稀释至1000 mL,再加三乙胺5 mL,混匀,用0.01 mol/L磷酸调节pH值至2.5)-甲醇(68:32);流速1.0 mL/min,检测波长277 nm,进样量10μL。结果氯霉素在60~160μg/mL范围内与峰面积呈良好的线性关系,回归方程为A=17.79C+17.13,r=0.9995;平均回收率为100.24%,RSD=1.26%。结论本方法简便快速,定量准确,结果稳定、重现性好,可用于氯霉素醇溶液中氯霉素的含量测定。

Objective To establish a method for the determination of Chloramphenicol in Chloramphenicol Alcoholic Solution by HPLC. Methods HPLC was used for quantitative analysis. Eclipse XDB-C18 (4.6 mmí250 mm, 5μm) was used, 0.01 mol/L heptane sodium sulfonate buffer solution (1000 mL 0.01 mol/L heptane sodium sulfonate buffer solu-tion consisted of 6.8 g dipotassium phosphate, 5 mL triethylamine and 0.01 mol/L heptanes sulfonic acid sodium solu-tion, adjusted pH to 2.5 by phosphoric acid solution) - methanol (68:32) was used as mobile phases, the flow rate was 1.0 mL/min, the detective wavelength was at 277 nm and the injection volume was 10 μL. Results The calibration curve was linear in the range of 60-160 μg/mL for Cholramphenicol, and the equation of linear regression was A=17.79C+17.13 (r = 0.9995); the average recovery rate was 100.24% and RSD was 1.26%. Conclusion The method is simple, accurate with good reproducibility and stability, and can be used for the quality control of the Chloramphenicol Alcoholic Solution.
