目的:研究同型半胱氨酸(HCY)对神经小胶质细胞(bv-2细胞)白介素-1β(IL-1β)、白介素-6(IL-6)表达的影响。方法:在细胞培养液中加入HCY、半胱氨酸(DL-CY)、空白对照组及溶剂对照组干预后,用细胞计数试剂盒(CCK8法)观察各组细胞增殖活性,并用酶联免疫吸附测定(ELISA)法测定细胞培养上清液IL-1β、IL-6浓度,用实时荧光定量核酸扩增检测系统(QPCR)的方法评价IL-1β、IL-6 mRNA表达改变。结果:实验各组中细胞增殖活性差异均无统计学意义(P>0.01),而HCY组IL-1β、IL-6的mRNA表达及上清液中蛋白表达与其他各组相比均有显著增加(P<0.01)。结论:HCY上调神经小胶质细胞IL-1β、IL-6的表达。

목적:연구동형반광안산(HCY)대신경소효질세포(bv-2세포)백개소-1β(IL-1β)、백개소-6(IL-6)표체적영향。방법:재세포배양액중가입HCY、반광안산(DL-CY)、공백대조조급용제대조조간예후,용세포계수시제합(CCK8법)관찰각조세포증식활성,병용매련면역흡부측정(ELISA)법측정세포배양상청액IL-1β、IL-6농도,용실시형광정량핵산확증검측계통(QPCR)적방법평개IL-1β、IL-6 mRNA표체개변。결과:실험각조중세포증식활성차이균무통계학의의(P>0.01),이HCY조IL-1β、IL-6적mRNA표체급상청액중단백표체여기타각조상비균유현저증가(P<0.01)。결론:HCY상조신경소효질세포IL-1β、IL-6적표체。
Objective:To investigate the effect of homocysteine on the expression of interleukin-1β(IL-1β), interleukin-6 (IL-6) in gitter cells. MethodThe BV-2 cells were cultured in the cell culture lfuid for 24 hours. HCY/DL-CY or the control solvent were added, and a cell counting kit (CCK8) was used to observe the proliferation activity. Enzyme linked immunosorbent assay (ELISA) was used to evaluate the concentration of IL-1βand IL-6 in the cell culture supernate. Well, mRNA levels of IL-1βand IL-6 were analysed by Real-time Quantitative PCR Detecting System (QPCR). ResultThe proliferation activity in each group were not signiif-cantly different, and the mRNA expression and protein expression of IL-1βand IL-6 in the supernatant homo-cysteine group was signiifcantly changed. Conclusion:Homocysteine increases the expression of IL-1β, IL-6 in gitter cells.
