目的:研究垂体肿瘤转化基因(PTTG)和血管生成与结直肠癌生物学行为的关系,探讨 PTTG 的表达与结直肠癌新生血管的关系。方法应用免疫组织化学法,检测120例结直肠癌、30例正常结直肠黏膜中 PTTG 的表达情况,并测定这些组织中微血管密度(MVD),结合临床病理学指标进行统计学分析。结果 PTTG 在结直肠癌患者中阳性表达率为84.17%,其阳性表达率显著高于对照组的6.67%( P ﹤0.01);其表达水平与患者 Dukes 分期及淋巴结转移有关( P ﹤0.05)。结直肠癌组织中 MVD 计数平均为34.75±5.15,显著高于对照组19.10±2.77( P ﹤0.01);并与结直肠癌的 Dukes分期、有无淋巴结转移有关( P ﹤0.05)。PTTG 表达阳性病例,其 MVD 计数普遍高于表达阴性病例;在表达阳性病例中, MVD 计数又与其阳性表达强度有关( P ﹤0.05)。结论 PTTG 表达及微血管密度与结直肠癌生物学特性关系密切;PTTG表达可能促进了结直肠癌肿瘤血管生成;联合检测 PTTG 和 MVD 可作为结直肠癌恶性程度及预后指标;抑制 PTTG 活性有可能为结直肠癌防治开辟新的前景。

목적:연구수체종류전화기인(PTTG)화혈관생성여결직장암생물학행위적관계,탐토 PTTG 적표체여결직장암신생혈관적관계。방법응용면역조직화학법,검측120례결직장암、30례정상결직장점막중 PTTG 적표체정황,병측정저사조직중미혈관밀도(MVD),결합림상병이학지표진행통계학분석。결과 PTTG 재결직장암환자중양성표체솔위84.17%,기양성표체솔현저고우대조조적6.67%( P ﹤0.01);기표체수평여환자 Dukes 분기급림파결전이유관( P ﹤0.05)。결직장암조직중 MVD 계수평균위34.75±5.15,현저고우대조조19.10±2.77( P ﹤0.01);병여결직장암적 Dukes분기、유무림파결전이유관( P ﹤0.05)。PTTG 표체양성병례,기 MVD 계수보편고우표체음성병례;재표체양성병례중, MVD 계수우여기양성표체강도유관( P ﹤0.05)。결론 PTTG 표체급미혈관밀도여결직장암생물학특성관계밀절;PTTG표체가능촉진료결직장암종류혈관생성;연합검측 PTTG 화 MVD 가작위결직장암악성정도급예후지표;억제 PTTG 활성유가능위결직장암방치개벽신적전경。
Objective To study the relationship between PTTG(Pituitary Tumor Transforming Gene),angiogenesis and biological behav-ior of colorectal cancer,as well as the relationship between the expression of PTTG and angiogenesis of colorectal cancer. Methods The expres-sion of PTTG and MVD had been examined in 120 patients with colorectal cancer and 30 samples of normal colorectal mucosa by immunohistochem-ical assay. The immunostaining results and related clinical - pathological data were analyzed by statistics. Results The positive expression rate of PTTG in colorectal cancer was 84. 17% ,and its positive expression rate was significantly higher than that of controls( P ﹤ 0. 01). Its positive ex-pression rate was related to Dukes staging and metastasis in lymph nodes of colorectal cancer( P ﹤ 0. 05). MVD in colorectal cancer was signifi-cantly higher than that of controls(34. 75 ± 5. 15 vs 19. 10 ± 2. 77,P ﹤ 0. 01). MVD was related to such factors as Dukes staging and lymphatic metastasis of colorectal cancer( P ﹤ 0. 05). MVD in cases with positive expression of PTTG was higher than that of cases with negative expression ( P ﹤ 0. 01),MVD was also correlated to the expression grade of PTTG( P ﹤ 0. 05). Conclusion The expression of PTTG and angiogenesis in colorectal cancer is significantly related to its biological characteristics,the expression of PTTG can cause the growth of vessels in colorectal cancer. Combined detection of PTTG and MVD can be used as indexes to determine the degree of malignancy and prognosis of colorectal cancer. The inhi-bition of activity of PTTG may bring new prospect in prevention and treatment of primary colorectal cancer.
