目的:观察芡实对糖尿病肾病大鼠肾组织中UrotensinⅡ及胶原表达的影响。方法:雄性SD大鼠60只,随机选取10只大鼠为正常对照组( N组),其余50只采用一次性腹腔注射链脲佐菌素( STZ,45 mg/kg)诱导DN模型,造模成功后随机分为糖尿病肾病组(DN)、芡实小剂量组(EL,1.5 g·kg-1·d-1)、中剂量组(EM,3.0 g·kg-1·d-1)、大剂量组(EH,6.0 g·kg-1·d-1)及氯沙坦钾组( LP,30 mg·kg-1·d-1),每组10只,均采用灌胃给药,N组和DN组给予等量蒸馏水。各组大鼠连续给药12周后,检测大鼠生化指标;HE、Masson染色观察肾脏病理变化;采用免疫组化、western-blot法测定Uro-tensinⅡ及胶原Ⅰ、Ⅲ在肾组织中表达的变化。结果:(1)与N组相比,12周末DN组大鼠肾小球肥大,系膜基质增生,间质胶原沉积,血清BUN、Scr、24 h尿蛋白定量明显增高(P<0.05),肾组织UrotensinⅡ及胶原Ⅰ、Ⅲ表达增加(P<0.05)。(2)与DN组相比,EM、EH组大鼠病理改变减轻,BUN、Scr、24 h尿蛋白定量显著下降( P<0.05),肾组织UrotensinⅡ及胶原Ⅰ、Ⅲ表达明显降低( P<0.05)。结论:芡实可通过抑制UrotensinⅡ及胶原Ⅰ、Ⅲ在肾间质的过度表达,进而改善肾功能,延缓糖尿病肾病大鼠肾间质纤维化的进程,从而发挥肾脏保护作用。

목적:관찰검실대당뇨병신병대서신조직중UrotensinⅡ급효원표체적영향。방법:웅성SD대서60지,수궤선취10지대서위정상대조조( N조),기여50지채용일차성복강주사련뇨좌균소( STZ,45 mg/kg)유도DN모형,조모성공후수궤분위당뇨병신병조(DN)、검실소제량조(EL,1.5 g·kg-1·d-1)、중제량조(EM,3.0 g·kg-1·d-1)、대제량조(EH,6.0 g·kg-1·d-1)급록사탄갑조( LP,30 mg·kg-1·d-1),매조10지,균채용관위급약,N조화DN조급여등량증류수。각조대서련속급약12주후,검측대서생화지표;HE、Masson염색관찰신장병리변화;채용면역조화、western-blot법측정Uro-tensinⅡ급효원Ⅰ、Ⅲ재신조직중표체적변화。결과:(1)여N조상비,12주말DN조대서신소구비대,계막기질증생,간질효원침적,혈청BUN、Scr、24 h뇨단백정량명현증고(P<0.05),신조직UrotensinⅡ급효원Ⅰ、Ⅲ표체증가(P<0.05)。(2)여DN조상비,EM、EH조대서병리개변감경,BUN、Scr、24 h뇨단백정량현저하강( P<0.05),신조직UrotensinⅡ급효원Ⅰ、Ⅲ표체명현강저( P<0.05)。결론:검실가통과억제UrotensinⅡ급효원Ⅰ、Ⅲ재신간질적과도표체,진이개선신공능,연완당뇨병신병대서신간질섬유화적진정,종이발휘신장보호작용。
Objective:To observe the effect of euryale ferox on expression of UrotensinⅡand collagen in diabetic nephrop-athy rats. Nethods:Sixty male Sprague-Dawley( SD)rats weighing 200~220 g were used. Randomly selected 10 rats in control group( group N),the remaining 50 making DN models injected streptozotocin into abdominal cavity( STZ,45 mg/kg),were random-ly divided into DN group(group DN),low-dose Euryale ferox group(group EL,1.5 g·kg-1·d-1),medium-dose Euryale fer-ox group(group EM,3.0 g·kg-1·d-1),high-dose Euryale ferox group(group EH,6.0 g·kg-1·d-1)and losartan potassium group( group LP,30 mg·kg-1 ·d-1 ),10 rats in each group. The rats of group EL,EM,EH and group LP were administrated by gavage. Rats of the other two groups were offered distilled water as much. Each group rats were administered continuously after 12 weeks,records of biochemical indicators and observes the renal pathological changes through HE and Masson dyeing. the immuno-histochemical and western blot methods were used to observe the expression of UrotensinII,collagenⅠ,Ⅲin renal tissues. Results:(1)As compared with Control group,DN group rats at the end of the 12 week,the glomerular of the rats became hypertrophic and me-sangial matrix increased,interstitial collagen deposition,24h urinary protein,serum BUN,Scr levels were significantly higher( P<0.05),U Ⅱ,collagen Ⅰ,Ⅲ expression increased(P<0. 05).(2)Compared with group DN,the pathology changes were re-lieved,BUN,Scr,24 h urinary protein as well as the expression of U Ⅱ,collagen Ⅰ,Ⅲ decreased in group EM and group EH (P<0. 05). Conclusion:Euryale ferox may inhibit overexpression of UⅡand the collagenⅠ,Ⅲin renal interstitial,improve kid-ney function,delay diabetic nephropathy tubulointerstitial fibrosis,and thus play a role in renal protection.
