Brassica rapa L.%Total flavonoids%Spectrophotometric,ultraviolet-visible
目的:通过单因素实验及正交实验探讨提取维药恰麻古中总黄酮的最佳工艺,并测定其不同产地的药材含量,为资源的合理开发利用提供科学依据。方法以乙醇为溶剂回流提取,采用紫外-可见分光光度法( UV)测定恰麻古中总黄酮的含量,并通过重复性、精密度和回收率实验考察实验方法。结果维药恰麻古中总黄酮的最佳工艺为乙醇浓度75%,提取时间2.5 h,提取温度80℃,料液比160。标准液及供试液均在510 nm波长处有最大吸收,总黄酮在0.008~0.048 mg· mL-1线性关系良好(r=0.9993),平均加样回收率99.59%;精密度实验、稳定性实验和重复性实验的RSD分别为0.022%,1.311%和1.601%,5个产地的恰麻古中,阿克苏恰麻古的黄酮含量最高,平均含量0.2802%。结论该方法操作简单,重复性好,灵敏度高,适用于恰麻古总黄酮的含量测定。

Objective To establish the optimum parameters for the flavonoids extraction from Brassica rapa L. by single-factor and orthogonal experiments,determine the content of flavonoids in Brassica rapa L. from different origins,and provide a scientific basis for the rational development and utilization of Brassica rapa L. Methods The total flavonoids in Brassica rapa L. was extracted by ethanol reflux extraction and the content was determined by ultraviolet-visible spectrophotometry. The method was validated by tests of stability, precision and recovery. Results The optimum extracting condition for flavonoids from Brassica rapa L. was as follows: using 75% ethanol as extracting agent,setting temperature at 80 ℃,using solid-liquid ratio at 1 g60 mL and extracting for 2. 5 h. The detection wavelength was 510 nm. The linear range and the detectable limit of the method were 0. 008-0. 048 mg· mL-1(r=0. 999 3). The average extraction recovery was 99. 59%. The RSD of the precision, stability and recovery tests were 0. 022%,1. 311%,and 1. 601%,respectively. The Brassica rapa L. from Akzo contained the highest flavonoids with an average value of 0. 280 2%. Conclusion This method is simple,reproducible,sensitive and can be used to determine the total flavonoids in Brassica rapa L . .
