目的:探讨exendin-4(Ex-4)预处理对大鼠缺血/再灌注(I/R)损伤的影响。方法雄性SD大鼠55只随机分为4组:假手术组(SO)( n =10),缺血/再灌注(I/R)组( n =15),Ex-4+I/R (Ex-4-I/R)组( n =15), Ex-4+exendin (9-39)[Ex-4-Ex(9-39)-I/R]组( n =15)。除SO组外,其余3组大鼠均行冠状动脉左前降支结扎,缺血30 min后再灌4 h,Ex-4-I/R组在I/R前给予Ex-4预处理,Ex-4-Ex(9-39)-I/R组在I/R前给予Ex-4和Ex(9-39)预处理。再灌注4h后进行心肌梗死面积评估,并取颈动脉血进行心肌酶、心肌氧化指标、心肌高迁移率族蛋白1(HMGB1)水平检测。结果缺血再灌注4h后,与I/R组比较,Ex-4-I/R组心肌梗死面积减少,分别为(54.1±3.0)%和(25.8±1.0)%,差异有统计学意义( P <0.05);与Ex-4-I/R组比较,Ex-4-Ex(9-39)-I/R组心肌梗死面积增大,为(47.8±3.0)%,差异有统计学意义( P <0.05)。与SO组比较,I/R组LDH、CK、MDA水平明显增加,HMGB1表达明显升高( P <0.05),而SOD水平明显下降( P <0.05);与I/R组比较,Ex-4-I/R组各项指标明显改善,差异有统计学意义(P <0.05);与Ex-4-I/R组比较,Ex-4-Ex(9-39)-I/R组中各项指标的改善均被抑制,差异有统计学意义( P <0.05)。结论 Ex-4预处理可能通过抑制HMGB1的表达进而减轻大鼠心肌I/R损伤。

목적:탐토exendin-4(Ex-4)예처리대대서결혈/재관주(I/R)손상적영향。방법웅성SD대서55지수궤분위4조:가수술조(SO)( n =10),결혈/재관주(I/R)조( n =15),Ex-4+I/R (Ex-4-I/R)조( n =15), Ex-4+exendin (9-39)[Ex-4-Ex(9-39)-I/R]조( n =15)。제SO조외,기여3조대서균행관상동맥좌전강지결찰,결혈30 min후재관4 h,Ex-4-I/R조재I/R전급여Ex-4예처리,Ex-4-Ex(9-39)-I/R조재I/R전급여Ex-4화Ex(9-39)예처리。재관주4h후진행심기경사면적평고,병취경동맥혈진행심기매、심기양화지표、심기고천이솔족단백1(HMGB1)수평검측。결과결혈재관주4h후,여I/R조비교,Ex-4-I/R조심기경사면적감소,분별위(54.1±3.0)%화(25.8±1.0)%,차이유통계학의의( P <0.05);여Ex-4-I/R조비교,Ex-4-Ex(9-39)-I/R조심기경사면적증대,위(47.8±3.0)%,차이유통계학의의( P <0.05)。여SO조비교,I/R조LDH、CK、MDA수평명현증가,HMGB1표체명현승고( P <0.05),이SOD수평명현하강( P <0.05);여I/R조비교,Ex-4-I/R조각항지표명현개선,차이유통계학의의(P <0.05);여Ex-4-I/R조비교,Ex-4-Ex(9-39)-I/R조중각항지표적개선균피억제,차이유통계학의의( P <0.05)。결론 Ex-4예처리가능통과억제HMGB1적표체진이감경대서심기I/R손상。
Objective To investigate the effect of exendin 4 ( Ex-4) preconditioning on rat ischemia/reperfusion ( I/R) damage.Methods Fifty-five male SD rats were randomly divided into 4 groups:sham operation group (SO) ( n =10), ischemia /reperfusion (I/R) group ( n =15),EX4+I/R (Ex-4-I/R) group (n=15), Ex-4+exendin (9-39)[Ex-4-Ex (9-39)-I/R] group ( n =15).Except SO group, the other 3 group rats underwent left anterior descending coronary artery ligation, after 30 min ischemia, reperfusion of 4 h, Ex-4-I/R group was given Ex-4 before I/R pretreatment, Ex-4-Ex(9-39)-I/R group was given Ex-4 and Ex-(9-39) preconditioning before I/R group.After reperfusion for 4h, then assessed of the my-ocardial infarction area , and get the carotid artery blood sample to test the myocardial enzyme , myocardial oxidative index , myocardial high mobility group protein 1 (HMGB1) levels.Results 4h after reperfusion, compared with group I/R, Ex-4-I/R group reduced myocardial infarct size more obviously , (54.1 ±3)% and (25.8 ±1)% respectively, the difference was statistically significant ( P <0.05) compared with Ex-4-I/R group, Ex-4-Ex(9-39)-I/R group myocardial infarction area in-creases, as of (47.8 ±3.0)%,the difference was statistically significant ( P <0.05).Compared with SO group, I/R group's LDH, CK, MDA levels were significantly increased, HMGB1 expression was significantly increased ( P <0.05), and SOD level decreased significantly ( P <0.05);compared with I/R group, Ex-4-I/R group's indicators improved significantly , the difference was statistically significant ( P <0.05);compared with the Ex-4-I/R group, Ex-4-Ex(9-39)-I/R group's indica-tors improvement were suppressed , the difference was statistically significant ( P <0.05).Conclusion It demonstrated that the Ex-4 preconditioning may inhibit the expression of HMGB 1 and reduce the myocardial I/R injury in rats .
