研究凋亡相关基因Bcl-2、Bax和caspase-3在金黄色葡萄球菌(S.aureus )感染人脐静脉内皮细胞株 ECV-304内的表达，探讨其对人脐静脉内皮细胞的凋亡作用。方法采用半定量反转录聚合酶链反应(RT-PCR)及免疫印迹(Western-blot)法检测S.aureus 感染后人脐静脉内皮细胞 Bcl-2、Bax和 caspase-3的 mRNA 及蛋白表达水平。结果 S.aureus 感染人脐静脉内皮细胞后，其细胞内与抗凋亡相关的细胞因子 Bcl-2 mRNA水平及蛋白表达量下降，感染12小时差异有统计学意义(P<0.01)，而促凋亡的细胞因子 Bax和 caspase-3表达上升，感染6小时差异有统计学意义(P<0.01)。结论凋亡相关细胞因子 Bcl-2、Bax和 caspase-3可能在S.aureus 感染所致的脐静脉内皮细胞凋亡中起重要的作用。
연구조망상관기인Bcl-2、Bax화caspase-3재금황색포도구균(S.aureus )감염인제정맥내피세포주 ECV-304내적표체，탐토기대인제정맥내피세포적조망작용。방법채용반정량반전록취합매련반응(RT-PCR)급면역인적(Western-blot)법검측S.aureus 감염후인제정맥내피세포 Bcl-2、Bax화 caspase-3적 mRNA 급단백표체수평。결과 S.aureus 감염인제정맥내피세포후，기세포내여항조망상관적세포인자 Bcl-2 mRNA수평급단백표체량하강，감염12소시차이유통계학의의(P<0.01)，이촉조망적세포인자 Bax화 caspase-3표체상승，감염6소시차이유통계학의의(P<0.01)。결론조망상관세포인자 Bcl-2、Bax화 caspase-3가능재S.aureus 감염소치적제정맥내피세포조망중기중요적작용。
Objective To study the expression of apoptosis related genes,Bcl-2,Bax and caspase-3 in human umbilical vein endothelial cells(ECV-304)infected with Staphylococcus aureus(S.aureus ),and to discuss how they regulate apoptosis of human umbilical vein endothelial cells.Methods The expression levels of apoptosis related genes were detected with semi-quantity reverse transcription-polymerase chain reaction(RT-PCR)and Western-blot.Results Cytokin Bcl-2 mRNA associated with anti-apoptosis and proteins decreased in the human umbilical vein endothelial cells infected with S.aureus for 1 2 hours,and that of Bax and caspase-3 increased significantly after 6 hour of the infection(P<0.01).Conclusion The apoptosis related genes,Bcl-2,Bax and caspase-3 may play important roles in regulation of apoptosis of the human umbilical vein endothelial cells infected with S.aureus .
